摘  要:以‘鲁林9号杨’无菌叶片与叶柄作为外植体,建立‘鲁林9号杨’的离体再生体系。结果表明,叶片诱导不定芽的最佳培养基为1/2 MS+0.3 mg·L^(-1)6-BA+0.15 mg·L^(-1)NAA,诱导出芽率为100%,平均出芽数为10.83个;叶柄诱导不定芽的最佳培养基为1/2 MS+0.5 mg·L^(-1)6-BA+0.1 mg·L^(-1)NAA,诱导出芽率为94%,平均出芽数为5.67个;不定芽生根的最佳培养基为1/2MS+0.15 mg·L^(-1)IBA+0.03 mg·L^(-1)NAA,其生根率为95.2%,平均每株生根5条。组培苗炼苗移栽成活率达95%。该研究旨在建立‘鲁林9号杨’的离体再生体系,为‘鲁林9号杨’品种遗传改良和分子生物学研究等提供试验基础,为培育高品质、速生、高抗的杨树新品种奠定理论基础。In this study,the sterile leaves and petioles of Populus deltoides×'Lulin No.9'were used to produce adventitious buds,then the adventitious buds were rooted to form regenerated plants.The results showed that the optimum medium of adventitious buds induced by the leaves of P.deltoides×'Lulin No.9'was 1/2 MS+0.3 mg·L^(-1)6-BA+0.15 mg·L^(-1)NAA,the induction rate of adventitious bud was 100%,and the average number of buds was 10.83.The optimum medium for inducing adventitious buds from petioles was 1/2 MS+0.5 mg·L^(-1)6-BA+0.1 mg·L^(-1)NAA,the induction rate of adventitious bud was 94%,and the average number of buds was 5.67.The best rooting medium was 1/2MS+0.15 mg·L^(-1)IBA+0.03 mg·L^(-1)NAA.The rooting rate was 95.2%,with an average of 5 roots per plant.The survival rate of the regenerated seedlings was 95%.The purpose of this study was to establish an in vitro regeneration system of P.deltoides×'Lulin No.9',to provide an experimental basis for genetic improvement and molecular biology research of P.deltoides×'Lulin 9',and to lay a theoretical foundation for cultivating new poplar varieties with high quality,fast growth and high resistance.
