摘 要:目的采用网络药理学方法探讨角药葛根-鹿衔草-羌活治疗颈椎病的作用机制。方法使用中药系统药理学数据库与分析平台筛选葛根、鹿衔草、羌活的主要有效活性成分及其作用靶点。采用CTD、GeneCards及OMIM数据库获取颈椎病的相关基因靶点,并将其与角药的有效活性成分作用靶点进行交集,获取共同靶点。针对共同靶点,构建“药物-成分-靶点-疾病”可视化网络,以及蛋白质-蛋白质互相作用(PPI)网络,同时进行基因本体论(GO)功能富集分析和京都基因和基因组百科全书(KEGG)通路富集分析。结果共获得葛根-鹿衔草-羌活的有效活性成分28个及其靶点86个,与12097个颈椎病相关基因靶点取交集后得到共同靶点72个。根据PPI网络筛选出白细胞介素6(IL-6)、半胱氨酸天冬氨酸蛋白酶3(Caspase3)、雌激素受体1(ESR1)等关键靶点蛋白。GO功能富集分析结果显示,共同靶点主要涉及核受体活性、转录因子活性/直接配体调节的序列特异性、类固醇激素受体活性等生物过程或功能;KEGG通路富集分析结果显示,葛根-鹿衔草-羌活治疗颈椎病的信号通路主要涉及丝裂原活化蛋白激酶(MAPK)信号通路、磷脂酰肌醇-3-激酶/蛋白激酶(PI3K/Akt)信号通路、细胞凋亡信号通路等多条通路。结论角药葛根-鹿衔草-羌活治疗颈椎病的作用,可能是其多种有效活性成分通过IL-6、ESR1等靶基因调控MAPK、PI3K/Akt等多条信号通路,并影响Caspase3的表达来实现的。Objective To explore the mechanism of action of horn medicine,Puerariae Lobatae Radix-Pyrolae Herba-Notopterygii Rhizoma et Radix,in the treatment of cervical spondylosis using network pharmacology method.Methods The Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform was employed to screen the main effective active ingredients and the action targets of Puerariae Lobatae Radix,Pyrolae Herba,and Notopterygii Rhizoma et Radix.CTD,GeneCards and OMIM databases were used to obtain the cervical spondylosis-related gene targets which were intersected with the action targets of horn medicine′s effective active ingredients to achieve common targets.In light of the common targets,a"drug-ingredient-target-disease"visual network as well as a protein-protein interaction(PPI)network was constructed,and Gene Ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed.Results A total of 28 effective active ingredients and 86 targets of Puerariae Lobatae Radix-Pyrolae Herba-Notopterygii Rhizoma et Radix were obtained.After the intersection between the aforementioned 86 targets and 12097 cervical spondylosis-related gene targets,72 common targets were obtained.According to the PPI network,key target proteins such as interleukin 6(IL-6),cysteinyl aspartate specific proteinase 3(Caspase3)and estrogen receptor 1(ESR1)were identified.The results of GO functional enrichment analysis showed that the common targets mainly involved biological processes or functions such as nuclear receptor activity,transcription factor activity/direct ligand-regulated sequence specificity and steroid hormone receptor activity;the results of KEGG pathway enrichment analysis revealed that the signaling pathways for cervical spondylosis treated by Puerariae Lobatae Radix-Pyrolae Herba-Notopterygii Rhizoma et Radix mainly involved multiple pathways such as mitogen-activated protein kinase(MAPK)signaling pathway,phosphoinositide-3-kinase/protein kinase B(P
