摘  要:环状RNA(circ RNA)由于其广泛性、稳定性、保守性等特点具有较高研究价值,已成为生物研究领域热点之一。为研究circ RNA在鸡脂肪细胞分化中作用,选择与前期发现在鸭脂肪细胞分化中具有重要作用的鸭circ_PLXNA1具有高度同源性的鸡circ_PLXNA1作验证分析。鸡circ_PLXNA1是一个稳定的环状转录本,核质分离试验发现鸡circ_PLXNA1主要定位在细胞质中。此外,circ_PLXNA1主要在鸡心脏和肝脏中表达。生物信息学分析结果表明,鸡circ_PLXNA1和预测的靶基因富集于Erb B、MAPK和Hedgehog等信号通路,表明鸡circ_PLXNA1可能通过调控靶基因间接参与脂肪细胞分化。Circular RNA(circRNA) has great research value due to its extensiveness, stability,conservation and other characteristics, and has become one of the hot spots in the field of biological research. In order to study the role of circRNA in the differentiation of chicken adipocytes, chicken circ_PLXNA1, which has a high degree of homology with duck circ_PLXNA1 and was found to play an important role in the differentiation of duck adipocytes, was selected for verification and analysis.Chicken circ_PLXNA1 is a stable circular transcript, and it was found that chicken circ_PLXNA1 was mainly located in the cytoplasm through nuclear and cytoplasmic separation experiments. In addition,circ_PLXNA1 was mainly expressed in the heart and liver of chickens. Bioinformatics analysis results showed that chicken circ_PLXNA1 and the predicted target genes were enriched in signaling pathways,such as ErbB signaling pathway, MAPK signaling pathway and Hedgehog signaling pathway, which revealed that chicken circ_PLXNA1 might play indirect function in adipocyte differentiation through the regulation of target genes.
