摘 要:为了筛选对甘薯蔓割病拮抗活性较好的菌株,采用平板对峙法对3株细菌菌株进行抑菌活性评估,并对新筛选的高效抑制甘薯蔓割病的菌株进行分子鉴定和紫外诱变研究.结果显示,细菌菌株KC2-8对尖孢镰刀杆菌有较强的抑制作用.通过16S rDNA序列检测后进行同源性分析,将该菌株鉴定为特基拉芽孢杆菌(Bacillus tequilensis).对KC2-8菌株进行紫外诱变显示,最佳诱变时间为20 s,进一步涂布筛选获得菌落直径大、生长迅速的6株稳定高产单菌株.综合分析表明,特基拉芽孢杆菌KC2-8对甘薯蔓割病菌具有良好抑制作用.To screen the bacteria strain with high antagonistic activity against sweetpotato stem rot,the inhibitory efficiencies of 3 strains of bacteria were determined by the methods of plate confrontation,as well as molecular identification and UV mutagenesis of the most efficient antagonistic strains were studied.The results reviewed that the culture filtrate of strain KC2-8 had the strongest inhibitory effect on sweetpotato stem rot.Based on 16 S rDNA sequence analysis,the strain KC2-8 was identified as Bacillus tequilensis.UV mutagenesis results showed that the optimal irradiation time of original stain of KC2-8 was 20 s.Through screening,6 stable mutant strain with high yield were obtain.This results indicated that Bacillus tequilensis KC2-8 had significant inhibitory effect on sweetpotato stem rot.
