摘 要:目的:探讨艾灸对快速老化小鼠(senescence-accelerated mouse prone/8,SAMP8)的学习记忆能力和海马突触超微结构的影响。方法:将18只SAMP8小鼠随机分为模型组和艾灸组,另取9只SAMR1小鼠为空白组,空白组、模型组小鼠不做干预,艾灸组小鼠艾灸关元穴。6周后用Morris水迷宫实验检测小鼠学习记忆能力的变化,透射电镜观察海马神经元突触形态并计算突触界面结构参数。结果:与空白组比较,模型组小鼠逃避潜伏期显著延长,穿越平台次数显著减少,目标象限游程比和停留时间比显著下降(P<0.05),电镜下可见突触超微结构损伤,突触间隙显著增宽,突触后致密物(postsynaptic density, PSD)厚度、突触界面曲率显著减小(P<0.05);与模型组比较,艾灸组逃避潜伏期显著下降,目标象限游程比显著上升(P<0.05),但目标象限停留时间比和穿越平台次数无显著性差异(P>0.05);电镜下突触形态结构改善,突触间隙宽度明显减小且PSD厚度显著增大(P<0.05),但突触界面曲率无显著性差异(P>0.05)。结论:艾灸干预能够缓解阿尔茨海默病小鼠海马区突触超微结构损伤,提高其学习记忆能力。Objective: To investigate the effect of moxibustion on the learning and memory ability and the ultrastructure of hippocampal synapses in senescence-accelerated mouse prone/8(SAMP8).Methods: Eighteen SAMP8 mice were randomly divided into model group and moxibustion group, and another 9 SAMR1 mice were selected as blank group.Mice in the blank group and model group were not intervened, while the mice in the moxibustion group were moxibustion at Guanyuan point.After 6 weeks, the Morris water maze test was used to detect the changes of the learning and memory ability of the mice, and the synaptic morphology of hippocampal neurons was observed by transmission electron microscope and the parameters of the synaptic interface were calculated.Results: Compared with the blank group, the escape latency of the model group was significantly prolonged, the number of crossing the platform was significantly reduced, and the target quadrant run ratio and dwell time ratio were significantly decreased(P<0.05).Compared with the model group, the escape latency of the moxibustion group was significantly decreased(P<0.05),and the target quadrant run ratio was significantly increased(P<0.05).However, there was no significant difference in the target quadrant residence time ratio and the number of crossing the platform(P>0.05);under the electron microscope, the synaptic morphology and structure were improved, the width of the synaptic gap was significantly reduced, and the thickness of the PSD was significantly increased(P<0.05),but there was no significant difference in the synaptic interface curvature(P>0.05).Conclusion: Moxibustion intervention can alleviate the synaptic ultrastructural damage in the hippocampus of Alzheimer′s disease mice and improve their learning and memory ability.
