治疗痛风中成药中掺杂西药成分吲哚美辛的含量测定

摘　　要：研究联合薄层色谱法-高效液相色谱法快速判定治疗痛风中成药中可能存在的化学药品成分.首先采用硅胶GF254薄层板,以醋酸泼尼松、吲哚美辛、醋酸地塞米松、布洛芬和秋水仙碱为对照品,乙酸乙酯:无水乙醇:氨水体积比50:16:1、氯仿:乙酸乙酯:冰醋酸体积比25:25:0.2、氯仿:乙酸乙酯:乙酸体积比30:10:1、饱和的乙酸乙酯和氯仿:甲醇体积比19:1为展开剂,初步确定抗痛风药丸中含有吲哚美辛.然后在色谱柱Eclipse XDB-C18(4.6×250 mm,5μm),流动相甲醇:冰醋酸(0.1 mol/L)体积比70:30,流速0.6 mL/min,检测波长320 nm的色谱条件下,有效地检测出该痛风中成药中吲哚美辛的含量,并对检测结果进行验证.结果显示,所用方法线性良好,具有较好的稳定性、重复性、精密度和回收率.The possible chemical components in Chinese patent medicines for Anti-gout pills by TLC-HPLC were detected and analyzed.First,under the silica gel GF254 thin plate,Prednisone acetate,Dexamethasone acetate,Indomethacin,Ibuprofen,and Colchicine were used as controls;and ethyl acetate:absolute ethanol:ammonia=50:16:1,chloroform:ethyl acetate:acetic acid=30:10:1,ethyl acetate:trichloromethane:glacial acetic acid=25:25:0.2,saturated ethyl acetate and chloroform:methanol=19:1 as developing solvent.Under this chromatographic conditions,Indomethacin in the Chinese traditional patent medicine for gout can be preliminarily detected.And the Eclipse XDB-C18(4.6×250 mm,5μm)column was used to determine the content of the mobile phase under the detection conditions of methanol:acetic acid(0.1 mol/L)=70:30,flow rate of 0.6 mL/min,and detection wavelength of 320 nm,and the content of indomethacin in Chinese gout was detected effectively.The results show that the method has good linearity,good stability,repeatability,precision and recovery.
