摘 要:目的观察不同剂量黄芩茎叶总黄酮对原发性高血压大鼠心肌重构的影响,并探讨其机制。方法将50只原发性高血压大鼠随机分为高血压组、贝那普利组及低、中、高剂量黄芩组,每组各10只,另取10只普通Wistar大鼠作为对照组。贝那普利组及低、中、高剂量黄芩组分别给予每日5.0 mg/kg贝那普利及17.5、35.0、70.0 mg/kg黄芩茎叶总黄酮灌胃处理,高血压组、对照组给予等量生理盐水灌胃。连续给药8周后,采用尾动脉容积法测定各组大鼠血压及心率;麻醉大鼠后分离左心室,计算各组大鼠心脏重量指数;HE染色法观察各组大鼠心肌组织形态;ELISA法检测各组大鼠心肌组织中心肌胶原指标Ⅰ型前胶原羧基末端肽(PⅠCP)、Ⅲ型前胶原氨基末端肽(PⅢNP)及核转录因子κB(NF-κB)表达。结果大鼠收缩压高血压组、低剂量黄芩组>中剂量黄芩组、高剂量黄芩组、贝那普利组>对照组(P均<0.05),各组大鼠心率比较差异均无统计学意义。大鼠心脏重量指数高血压组、低剂量黄芩组>中剂量黄芩组、高剂量黄芩组、贝那普利组>对照组(P均<0.05)。对照组大鼠心肌组织间隙较窄,可见少量胶原占据;高血压组大鼠心肌组织间隙增宽,肌间隙中胶原含量增多;中、高剂量黄芩组及贝那普利组大鼠心肌组织较高血压组间隙变窄且胶原减少;低剂量黄芩组较高血压组心肌结构改善不明显。大鼠心肌组织PⅠCP、PⅢNP比较,高血压组、低剂量黄芩组高于中剂量黄芩组、高剂量黄芩组、贝那普利组、对照组(P均<0.05);大鼠心肌组织NF-κB表达比较,高血压组、低剂量黄芩组>中剂量黄芩组、高剂量黄芩组、贝那普利组>对照组(P均<0.05)。结论黄芩茎叶总黄酮可抑制原发性高血压大鼠的心肌重构,且中剂量应用时效果最好,其机制可能与抑制NF-κB信号通路引发的炎症反应有关。Objective To observe the effects of different doses of Huangqin(Scutellaria Baicalensis Georgi)on myocardial remodeling in rats with primary hypertension,and to investigate their possible mechanisms.Methods Fifty spontaneously hypertensive rats(SHR)were randomly divided into the hypertension group,benazepril group,and low-dose,medium-dose,and high-dose Huangqin groups,with 10 rats in each group.Ten normal Werster rats were used as the control group.Rats in the benazepril group,and low-dose,medium-dose,and high-dose Huangqin groups were given benazapril(5.0 mg/kg),and 17.5,35.0,and 70.0 mg/kg total flavonoids from stems and leaves of Huangqin,respectively.Rats in the hypertension group and control group were given equal volume of normal saline.After 8 weeks of treatment,blood pressure and heart rate were measured by caudal artery volume method.After anesthesia,the left ventricle was separated and the cardiac weight index was calculated.The expression levels of type Ⅰ procollagen carboxy-terminal peptide(PⅠCP),typeⅢprocollagen N-terminal peptide(PⅢCP)and nuclear transcription factorκB(NF-κB)were detected by ELISA.Myocardial histomorphology of rats in each group was observed by HE staining.Results The systolic pressure was the highest in the hypertension group and low-dose Huangqin group,and was the lowest in the control group(all P<0.05).There was no significant difference in the heart rate among five groups(P<0.05).The cardiac weight indices of rats in the hypertension group and low-dose Huangqin group were higher than those in the medium-dose and high-dose Huangqin groups and benazepril group,and were the lowest in the control group(all P<0.05).PⅠCP and PⅢCP in the myocardial tissues were higher in the hypertension group and low-dose Huangqin group than in the medium-dose Huangqin group,high-dose Huangqin group,benazepril group,and control group(all P<0.05).The expression of NF-κB in the myocardial tissues of rats in the hypertension group,low-dose Huangqin group was higher than that in the medium-
