丁酸对大肠杆菌生长曲线的影响

摘　　要：本试验主要研究丁酸对大肠杆菌生长曲线的影响,并用醋酸进行对比试验,为丁酸代替饲用抗生素的研究提供一定的依据。采用牛津杯法检测丁酸、醋酸(0.125%~4%)对大肠杆菌的抑菌效果;并使用酶标仪测定细菌在LB肉汤(0.025%~0.1%)中0~24 h的OD600值,根据相应数值绘制大肠杆菌的生长曲线。在牛津杯法中,4%丁酸、醋酸抑菌效果最好,抑菌圈直径分别达22 mm、28 mm;浓度为0.5%时,抑菌圈直径仅为9 mm、10 mm;浓度为0.25%、0.125%时,没有出现明显的抑菌圈。酶标仪测OD值法中,丁酸、醋酸浓度为0.1%时,大肠杆菌经过24 h检测其OD_(600)值基本不变;浓度为0.075%的OD值在19 h才有轻微增长。丁酸和醋酸对大肠杆菌生长均有抑制作用,浓度越高对大肠杆菌的生长抑制作用越大。This experiment mainly studied the effect of butyric acid on the growth curve of Escherichia coli,and carried out a comparative experiment with acetic acid,so as to provide a basis for the study of butyric acid instead of feed antibiotics.Oxford cup method was used to detect the antibacterial effect of butyric acid and acetic acid(0.125%~4%)on Escherichia coli;The OD value of bacteria in LB broth(0.025%~0.1%)for 0~24 h was determined by microplate reader(OD600),and the growth curve of E.coli was drawn according to the corresponding values.In Oxford cup method,the concentration of the butyric acid and acetic acid,which was 4%,had the best bacteriostatic effect,and the diameter of bacteriostatic circle reached 22 mm and 28 mm;The concentration of 0.5%had the worst bacteriostatic effect,and the bacteriostatic diameter was only 9 mm and 10 mm;At the concentration of 0.25%and 0.125%,there was no obvious inhibition zone.In the method of OD value measured by microplate reader,the OD_(600) value of Escherichia coli with butyric acid and acetic acidconcentration of 0.1%remained basically unchanged after 24 hours,and the OD value with concentration of 0.075%increased slightly after 19 hours.Butyric acid and acetic acid have inhibitory effects on the growth of Escherichia coli.The higher the concentration,the greater the inhibitory effect on the growth of Escherichia coli.
