摘  要:本试验旨在探讨不同剂量环磷酰胺对福建黄兔肝肾组织造成的损伤程度。选择雌性成年福建黄兔18只,随机分为对照组(生理盐水),剂量组(Ⅰ~Ⅴ,环磷酰胺浓度分别为20 mg/kg、40 mg/kg、60 mg/kg、80 mg/kg、100 mg/kg),每组3只,连续腹腔注射7 d,第8 d处死动物并解剖取肝脏、肾脏组织,制备肝肾组织石蜡切片,光学显微镜检查组织学结构变化。结果显示,低剂量(20 mg/kg)环磷酰胺对福建黄兔肝肾组织形态无明显影响,高剂量(80 mg/kg及以上)可诱导福建黄兔肝细胞变性,严重者坏死,同时可导致肾小管上皮变性,肾间质水肿、渗出,肾小管上皮细胞坏死脱落,肾小球缺血萎缩。二者病变均随剂量增加而加重。研究表明,不同剂量环磷酰胺造成福建黄兔肝脏、肾脏组织不同程度的损伤。The aim of this study was to investigate the injury effect of different doses of cyclophosphamide on liver and kidney tissue of Fujian yellow rabbits.Eighteen adult female Fujian yellow rabbits were randomly divided into 6 groups,injected intraperitoneally with 20 mg/kg,40 mg/kg,60 mg/kg,80 mg/kg,100 mg/kg cyclophosphamide for 7 consecutive days in five dose groups(cyclophosphamide I to V),respectively,except the control group(normal saline).The liver and kidney tissues were collected on the 8th day.Paraffin sections of liver and kidney tissue were prepared and the histological changes were observed by optical microscopy.The results showed that low-dose(20 mg/kg)cyclophosphamide had no significant effect on the liver and kidney tissue of Fujian yellow rabbits.High doses(80 mg/kg and above)cyclophosphamide could induce hepatocyte degeneration,and then necrosis in severe cases.Kidney tissue damages were mainly manifested in renal tubular epithelial degeneration,renal interstitial edema and exudation,necrosis and shedding of renal tubular epithelial cells,glomerular ischemia and atrophy.Both lesions aggravated with increasing dose.The studies showed that different doses of cyclophosphamide caused different degrees of liver and kidney tissue damage in Fujian yellow rabbits.
