摘  要:【目的】评价甘蔗野生种质资源的遗传多样性,为甘蔗抗逆定向遗传改良挖掘具有育种潜力的种质资源。【方法】对44份甘蔗野生近缘种材料按照割手密种内、斑茅种内以及种间3个层次运用ISSR进行遗传多样性分析。【结果】44个种间材料利用13条ISSR引物扩增,获得261条条带,其中259条为多态性条带、占比为99%;种间多态性依次为甘蔗属割手密种>甘蔗属斑茅种>蔗茅属蔗茅种>芒属五节茅种>金茅属金茅种>河八王属河八王种;44份材料遗传相似系数为0.125~0.710,UPGMA聚类结果基本同形态学分类一致,12个斑茅种样品在0.805处聚合,斑茅和割手密在0.775处聚合,采自西藏察隅的编号A30、A32蔗茅关系密切,芒属、蔗茅属、河八王属的亲缘关系更近,与甘蔗属材料在0.73处聚合。【结论】甘蔗野生种质资源具有较高的遗传多样性,为甘蔗种质资源创新、遗传育种提供了种质材料和理论依据。[Objective]The purpose of the present paper is to evaluate the genetic diversity of wild sugarcane germplasm resources,and to explore germplasm resources with breeding potential for directional genetic improvement of sugarcane stress resistance.[Method]In the study,the genetic diversity of 44 wild relatives materials divided into three levels according to inter-species,Saccharum spontaneum,S.arundina-ceus were analyzed by ISSR molecular markers.[Result]Using 13 ISSR primers screened in 44 inter-species materials,a total of 261 DNA fragments were amplified,among which 259(99%)were polymorphic;Interspecies polymorphism:Saccharum spontaneum>Saccharum arundinaceus>Erianthus fulvus>Miscanthus floridulus>Eulalia speciosa>Narenga porphyrocoma;The genetic similarity coefficient a-mong 44 inter-species materials varied from 0.125 to 0.710.The UPGMA clustering results of the 44 interspecies materials were basically consistent with the morphological classification.The 12 S.arundinaceus materials were clustered together in 0.815;S.spontaneum and S.arundinaceus materials were close cluster together in 0.75,Narenga;Miscanthus,Erianthus materials were closer than others clustering in 0.73 with Saccharum materials.[Conclusion]The study showed that sugarcane wild germplasm resources had high genetic diversity,which could provide germplasm materials and theoretical basis for sugarcane genetic breeding.
