摘 要:【目的】研究贵州毕节地区四川裂腹鱼(Schizothorax kozlovi)养殖群体的遗传多样性,为其选育和遗传改良工作提供理论依据。【方法】PCR扩增、DNA测序获取四川裂腹鱼养殖群体D-loop区和Cyt b基因序列,采用相关软件分析序列变异和群体遗传多样性。【结果】四川裂腹鱼养殖群体D-loop区和Cyt b基因序列分别检测到9、34个核苷酸多态位点(S),对应定义7和8种单倍型,单倍型多样性(Hd)和核苷酸多样性(π)分别为0.765、0.842和0.00519、0.00826;中性检测和核苷酸错配分析显示四川裂腹鱼养殖群体近期历史上可能未经历群体扩张事件。【结论】毕节地区四川裂腹鱼养殖群体具有较为丰富的遗传多样性,四川裂腹鱼养殖群体选择压力主要受到负向选择的影响,当前的人工选择对其Cyt b基因进化的影响有限。【Objective】In order to supply theoretical basis for the resource protection and genetic improvement of Schizothorax kozlovi, the genetic diversity of one cultured population from Bijie, Guizhou was assessed by mitochondrial control region(D-loop) segments and Cyt b gene sequences. 【Method】The partial sequences of D-loop region and Cyt b gene in farmed S. kozlovi were obtained by PCR and DNA sequencing, then sequence variation and genetic diversity of the two sequences were analyzed using related softwares. 【Result】9 and 34 polymorphic loca, as well as 7 and 8 haplotypes were detected in the sequences of D-loop region and Cyt b gene respectively, the haploidtype diversity(Hd) and nucleotide diversity(π) were 0.765, 0.842 and 0.00 519, 0.00 826 respectively, on the basis of the sequences of D-loop region and Cyt b gene in the population. Analysis of neutral detection and nucleotide mismatch distribution showed that there might be no expansion ocurred recently in the population of S. kozlovi. 【Conclusion】High genetic diversity was observed in the farmed S. kozlovi from Bijie, Guizhou. The result of selective pressure tests infered that negative selection played a positive role in the Cyt b gene of farmed S. kozlovi,that was, artificial selection served limited functions.
