摘 要:目的:探讨PM_(2.5)混悬液对人肺癌A549细胞、人肝癌HepG2细胞和人肾癌A498细胞迁移和侵袭能力的影响。方法:采用10、50μg/mL的PM_(2.5)混悬液分别对A549、HepG2和A498细胞染毒24 h,Transwell小室实验检测细胞迁移、侵袭能力。结果:迁移实验结果显示,阴性对照组、10和50μg/mL PM_(2.5)染毒组A549细胞穿膜细胞数分别为(356.33±30.92)、(476.33±28.50)和(563.33±74.00)个;HepG2细胞分别为(86.42±4.75)、(140.00±16.15)和(234.17±9.06)个;A498细胞分别为(42.33±6.43)、(80.00±6.08)和(174.67±25.01)个。侵袭实验结果显示,阴性对照组、10和50μg/mL PM_(2.5)染毒组A549细胞的穿膜细胞数分别为(188.33±9.45)、(313.00±7.94)和(404.00±57.66)个;HepG2细胞分别为(48.33±3.26)、(97.75±6.63)和(123.92±8.57)个;A498细胞分别为(5.67±5.03)、(21.33±5.13)和(40.67±6.81)个。在迁移实验和侵袭实验中,与阴性对照组相比,PM_(2.5)染毒组中的3种细胞穿膜细胞数均增加(P<0.05或0.01)。结论:PM_(2.5)可增强A549、HepG2和A498细胞的迁移和侵袭能力。OBJECTIVE:To investigate effects of PM_(2.5) on the migration and invasion abilities in three types of cancer cells:human lung(A549),human liver(HepG2)and human renal(A498)cancer cells.METHODS:The A549,HepG2 and A498 cells were exposed to PM_(2.5) suspension at 10 and 50μg/mL for 24 h.The transwell assay was performed to detect cell migration and invasion abilities.RESULTS:The results from the migration assay show that the number of transmembrane A549 cells in the negative control,10 and 50μg/mL PM_(2.5) exposure groups were(356.33±30.92),(476.33±28.50)and(563.33±74.00),respectively.Data for the HepG2 cells were(86.42±4.75),(140.00±16.15)and(234.17±9.06);and those for the A498 cells were(42.33±6.43),(80.00±6.08)and(174.67±25.01),respectively.The results from the invasion assay show that the number of transmembrane A549 cells in the three experimental groups were(188.33±9.45),(313.00±7.94)and(404.00±57.66),respectively.Data for the HepG2 cells were(48.33±3.26),(97.75±6.63)and(123.92±8.57);and those for the A498 cells were(5.67±5.03),(21.33±5.13)and(40.67±6.81),respectively.In the migration and invasion experiments,compared with the negative control group,the numbers of penetrating cells of the three types of cells in the PM_(2.5) exposure group were significantly increased(P<0.05).CONCLUSION:PM_(2.5) significantly enhanced the abilities of migration and invasion in A549,HepG2 and A498 cells.
