摘 要:为鉴定一起花边鸭突然发病死亡的病原,从发病鸭中分离到一株致病菌GZZY2019,分离菌在鲜血培养基中呈现圆形、表面光滑、边缘规则、半透明的露滴状小菌落;革兰氏染色镜检显示,分离菌为革兰氏阴性短杆菌;生化鉴定结果显示,分离菌能发酵葡萄糖、甘露醇等,鸟氨酸、尿素、吲哚等试验阳性;细菌种特异性鉴定结果与多杀性巴氏杆菌相符;荚膜分型鉴定结果仅扩增出约1050bp的基因片段,与荚膜血清A型相符;动物回归试验显示,该分离菌具有较强的致病性;16s rDNA基因序列比对显示该分离菌与多杀性巴氏杆菌LSBS1(GenBank登录号:MN080875.1)同源性高达99.93%;毒力基因检测结果显示该菌能检出kmt1、hgbA、pfhA、nanB、ptfA等毒力因子;药敏试验结果显示,GZZY2019对头孢他啶、丁胺卡那及氯霉素等6种药物耐药。To identify the pathogen of a sudden illness and death of lace ducks,a strain of pathogenic bacteria named G22Y2019 was isolated.It shows round,smooth surface,regular edges and translucent dew-shaped colonies in the fresh blood culture medium;Gram stain microscopic examination showed that the isolate was Gram-negative brevis;the biochemical identification results showed that the isolate could ferment glucose,mannitol,etc.,and the tests of ornithine,urea,indole,etc.were positive;the bacterial species-specific identification results were consistent with Pasteurella multocida;the capsular typing identification results only amplified a gene fragment of about 1050 bp,which was consistent with the capsular serotype A.The animal regression test showed that the isolate had strong pathogenicity;the 16s rDNA gene sequence alignment showed that the isolate had a homology of up to 99.93%with Pasteurella multocida LSBS1(GenBank accession number:MN080875.1);the virulence gene test results showed that the bacterium can detect kmt1,hgbA,and pfhA,NanB,ptfA and other virulence factors;drug sensitivity test results showed that it is resistant to 6 drugs including Ceftazidime,Amikacin and Ahloramphenicol.
