摘  要:从1年生及4年生纳塔栎母树上剪取插穗,在吲哚丁酸(IBA)、萘乙酸(NAA)、间苯三酚(THB)混合溶液中处理1 h,三者的质量浓度分别为500、250、50 mg·L^(-1),以清水处理相同时间为对照(CK),测定各处理插穗的愈伤率、生根率、一级根数、平均根长,同时研究了8、9、11月3个不同采穗时期纳塔栎生根率的差异,并采用GC-MS技术对插穗生根过程中影响其生根的物质相对百分比进行了测定。结果表明:植物激素对纳塔栎扦插生根有显著影响,1年生、4年生母树的插穗均是处理组的生根率高于对照组。母树年龄、采穗时期对纳塔栎生根也有一定影响,其中,从1年生母树剪取的插穗,各生根指标均高于4年生母树的插穗。对于不同采穗时期,1年生母树的插穗在8月份扦插的生根率较9月份略高,但二者差异不显著,11月份扦插的生根率显著下降至20.2%;4年生母树的插穗,3个时期扦插的生根率差异显著,其中以8月份最高,11月份最低。在生根过程中,对影响纳塔栎插穗生根的物质进行GC-MS测定,发现γ-谷甾醇、桦木脑、正癸酸、邻苯三酚等物质,其中γ-谷甾醇、桦木脑在生根过程中相对百分比呈现上升趋势;正癸酸的相对百分比则呈现下降的变化趋势;邻苯三酚呈先下降后上升的趋势。选取1年生纳塔栎苗木,在8月份剪取枝条进行扦插,其生根效果最好。经GC-MS鉴定,初步推断纳塔栎插穗提取液中γ-谷甾醇、桦木脑为促进扦插生根的物质,正癸酸为抑制生根的物质。To provide theoretical basis and technical reference for the propagation of Quercus nuttallii cuttings,the cuttings from the parent stocks of 1-and 4-year-old were treated with a combined solution of concentrations of IBA,NAA and THB were 500,250 and 50 mg·L^(-1) for 1 h.Cuttings were treated for the same duration with water as the control(CK).The indexes of callus rate,rooting percentage,primary root number and average root length of cuttings were measured.Meanwhile,the rooting effects of Q.nuttallii was studied in three different cutting times of August,September and November.The relative percentage of substances during the rooting process were determined by GC-MS.The plant hormones had a significant effect on the rooting percentage of Q.nuttallii.The rooting percentage of treated groups from the parent stock of 1-and the 4-year-old were higher than the control.The rooting percentage of Q.nuttallii was influenced by the age of stock plants and the seasons of cuttings.Among them,the rooting parameters from 1-year-old seedlings were higher than those from 4-year-old trees.For different cutting periods,the rooting percentage from 1-year-old tree in August was slightly higher than that in September,but there was no significant difference between them.The rooting percentage in November of 20.2%was significantly lower.The rooting percentage of cuttings from 4-year-old mother trees was significantly different in the three periods,among them the highest rooting percentage was in August and the lowest was in November.During the rooting process,the substances ofγ-sitosterol,betulin,n-decanoic acid and pyrogallic acid were determined by GC-MS,and the relative percentage ofγ-sitosterol and betulin showed the increasing trends,whereas the relative content of n-decanoic acid indicated a decreasing trend.Additionally,the relative percentage of pyrogallic acid showed a trend of decreasing first and then increasing.In summary,the results showed that 1-year-old Q.nuttallii parent stocks were suitable for cutting in August owi
