摘  要:丙酮酸激酶(pyruvate kinase,PYK)是昆虫能量代谢过程中的一个关键酶.为探究该基因调控异色瓢虫(Harmonia axyridis)生殖力的功能,采用RNA干扰技术抑制PYK1-1基因的表达,测定产卵前期、30 d总产卵量、孵化率及关键基因的表达水平,并观察卵巢发育情况.将dsPYK1-1导入异色瓢虫成虫体内后,雌虫的产卵前期延长、前20 d总产卵量低于未注射组和注射ds GFP组.相较于未注射组和注射ds GFP组,dsPYK1-1组卵巢发育较迟缓、体型稍小,而卵的孵化率无显著变化.雌虫卵黄原蛋白基因Vg1的表达水平在注射后第3天显著高于ds GFP组,Vg1与Vg2的表达水平在注射后第5天和第7天均显著低于ds GFP组;雌虫卵黄原蛋白受体基因VgR在注射后第5天显著低于ds GFP组.结果表明:PYK1-1能通过能量代谢影响雌虫卵巢发育和Vg与VgR表达,进而调控异色瓢虫生殖力.Pyruvate kinase(PYK)is a key enzyme in the process of insect energy metabolism.In order to explore the function of PYK in regulating the fecundity of Harmonia axyridis,RNA interference technology was used to inhibit the expression level of PYK1-1 gene.Then,the pre-oviposition,30-day egg production,hatching rate and the expression levels of key genes were measured.In addition,the ovarian development was also observed.The results showed that after ds PYK1-1 was injected into the adult of Harmonia axyridis,the pre-oviposition period of the females was prolonged,and the total number of eggs laid within first 20 days was significantly lower than that in non-injected group and ds GFP-injected group.The ovarian development of the ds PYK1-1 treatment was slower and the size was slightly smaller than other treatments,but the hatching rate of eggs had no significant differences.Moreover,the expression level of female vitellogenin Vg1 was significantly higher than that of the ds GFP group on the 3rd day after injection,while the expression levels of Vg1 and Vg2 were significantly lower than those of the ds GFP group on the 5th and 7th day after injection.The vitellogenin receptor VgR of female was significantly lower than in ds GFP group on the 5th day after injection.The results indicate that PYK1-1 can affect female ovarian development and the expression of Vg and VgR through energy metabolism,thereby regulating the fertility of H.axyridis.
