基于多指标成分和药效学试验优选芪丹益肺通窍方的提取工艺

摘　　要：目的:优选芪丹益肺通窍方的提取工艺。方法:以10种成分含量及干膏率为指标,采用正交试验法优选加水量、提取时间、提取次数等工艺参数;采用大鼠过敏性鼻炎模型评价芪丹益肺通窍方水提物的疗效。结果:与模型组比较,芪丹益肺通窍方工艺提取物可明显减轻过敏性鼻炎大鼠模型的鼻痒、喷嚏、流涕症状,减少鼻黏膜渗出物与炎症细胞产生,升高血清IL-10和降低IgE水平差异有统计学意义(P<0.05)。最佳提取工艺为加5倍量水,提取3次,每次0.5 h。结论:芪丹益肺通窍方水提物在减轻过敏性鼻炎大鼠模型症状、改善鼻黏膜组织形态学及降低免疫因子水平方面疗效确切,表明优选的芪丹益肺通窍方水提工艺稳定、可行,为该方后续的新制剂研究提供依据。OBJECTIVE Optimize the extraction process of Qidan Yifei Tongqiao Recipe.METHODS Optimize the process parameters, such as water addition, extraction time and extraction times by orthogonal test through to the contents of 10 components and dry paste rate as indexes.Evaluate the effect of water extract of Qidan Yifei Tongqiao Recipe by model of allergic rhinitis in rats.RESULTS Compared with the model group, the process extract of Qidan Yifei Tongqiao Recipe could significantly reduce the symptoms of nasal itching, sneezing and runny nose in the rat model of allergic rhinitis, reduce the production of nasal mucosal exudates and inflammatory cells, and increase the level of serum IL-10 and decrease the level of IgE with statistical significance(P<0.05).The best extraction process was to add 5 times of water, extract 3 times, 0.5 hour each time.CONCLUSION The water extract of Qidan Yifei Tongqiao Recipe has a definite effect on alleviating the symptoms of allergic rhinitis in rats, improving the histology of nasal mucosa and reducing the level of immune factors.The results showed that the optimal water extraction process of Qidan Yifei Tongqiao Recipe was stable and feasible, which provide a basis for the follow-up study of new preparations.
