摘 要:目的:筛选兰香草药材中总黄酮和木犀草素最佳超声提取工艺,并探究其在不同采收期的含量动态变化趋势,寻找最佳采收期。方法:正交试验结合单因素考察筛选出超声提取兰香草中总黄酮和木犀草素的最佳工艺,采用紫外-可见分光光度法测定不同采收期兰香草中总黄酮含量,采用高效液相色谱法测定不同采收期兰香草中木犀草素含量,分析其含量动态变化规律。结果:最佳超声提取工艺条件为:乙醇浓度为70%,料液比为1∶40,提取时间为60min,提取次数为2次。随着兰香草药材植株生长,总黄酮和木犀草素含量总体变化趋势均是先不断递增,9月达到峰值,之后逐渐减低;其中,木犀草素含量8月与9月无显著性差异(P>0.05)。结论:该提取工艺简单合理、稳定可行,可用于兰香草中总黄酮和木犀草素的提取。兰香草药材采收,以总黄酮含量为评价指标时,9月采收最佳;以木犀草素含量为评价指标时,8月和9月采收最佳。To screening the best ultrasonic extraction technology of total flavonoids and luteolin from Caryopteris incana,and to explore the dynamic trend of their content of Caryopteris incanain different harvest periods to find the best harvesting period.Methods:Orthogonal experiment combined with single-factor investigation was used for screening out the best technology for ultrasonic extraction of total flavonoids and luteolin of Caryopteris incana.The content of total flavonoid of Caryopteris incanain different harvest periods was determined by ultraviolet-visible spectrophotometry,the content of luteolin of Caryopteris incanain different harvest periods was determined by high performance liquid chromatography,and the dynamic change law of their contents was analyzed.Results:The optimal ultrasonic extraction process conditions as follows:ethanol concentration was 70%,material-to-liquid ratio was 1∶40,extraction time was 60 min,and extraction times was 2 times.With the growth of Caryopteris incana plants,the overall trend of the total flavonoids and luteolin content was increased first,reached a peak in September,and then decreased gradually.The content of total flavonoids and luteolin of Caryopteris incanain different harvesting periods wasdifferent.Among them,the content of luteolin was no significantly different from August and September(P>0.05).Conclusion:The extraction process was simple,reasonable,stable and feasible.And it can be used for the extraction of total flavonoids and luteolin of Caryopteris incana.For the harvest of Caryopteris incana,when the total flavonoid content was used as the evaluation index,the best harvest in September.When the luteolin content was used as the evaluation index,the best harvest in August and September.
