摘 要:磷素是植物的生长发育所必需的元素之一。为探究甜菜应对低磷胁迫吸收转运磷的机制,挖掘甜菜磷酸盐转运蛋白基因,采用同源克隆技术从甜菜克隆到两个编码磷酸盐转运蛋白的基因,命名为BvPT1;4和BvPHO1。生物信息学分析表明BvPT1;4蛋白编码526个氨基酸残基,蛋白分子量为57859.43 Da,等电点为8.76,是一个定位在细胞质膜上的疏水、无信号肽的非分泌蛋白,具有高亲和力磷酸盐转运蛋白的跨膜结构。BvPHO1蛋白编码797个氨基酸残基,蛋白分子量为91910.50 Da,等电点为9.24,是一个定位在细胞质膜上的亲水、无信号肽的非分泌蛋白,具有7个跨膜结构。进化分析结果表明,甜菜BvPHO1、菠菜SpoPHO1和藜麦CqPHO1的同源关系近,BvPT1;4、菠菜SpoPT1;4和藜麦CqPT1;7的同缘关系最近。研究结果进一步为甜菜优良品种的培养以及开展甜菜磷酸盐转运蛋白的研究奠定了基础。Phosphorus is one of the essential macronutrient for plant growth.In order to explore the mechanism of phosphorus uptake and transport in sugar beet in response to low phosphorus stress,two phosphate transporter genes were cloned from sugar beet by homologous cloning technique,named as BvPT1;4 and BvPHO1.Bioinformatics analysis showed that BvPT1;4 protein encodes 526 amino acid residues,molecular weight of protein 57859.43 Da,isoelectric point of 8.76.It is a hydrophobic,non-secreted protein without signal peptide located on the cell plasma membrane,transmembrane structure of phosphate transporter with high affinity. BvPHO1protein encodes 797 amino acid residues with a molecular weight of 91910.50 Da and an isoelectric point of 9.24.It is a hydrophilic,non-secreted protein without signal peptide and has 7 transmembrane structures located on the plasma membrane of cells.Evolutionary analysis showed that sugar beet BvPHO1,spinach SpoPHO1 and quinoa CqPHO1 are closed-relatived homology.BvPT1;4 and spinach SpoPT1;4 and Quinoa CqPT1;7are more closely-related hymology.These results laid a foundation for the cultivation of superior varieties of sugar beet and the study of phosphate transporters in sugar beet.
