摘 要:为探究过量铁对绵羊脾脏主要炎症因子和组织病理学的影响,将16只美利奴杂交公绵羊随机分为四组:对照组(CON)、铁过量组1(IronE1)、铁过量组2(IronE2)和铁过量组3(IronE3)。四组分别在基础日粮中添加50、500、1000和1500 mg/kg的一水合硫酸亚铁(FeSO;·H;O),饲养75 d后宰杀,取出脾脏。采用荧光定量PCR技术检测绵羊脾脏白细胞介素-1β(interleukin-1,IL-1β)、白细胞介素-2(interleukin-2,IL-2)、白细胞介素-6(interleukin-6,IL-6)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、干扰素-γ(interferon-γ,IFN-γ)、核因子-κB(nuclear factor-κB,NF-κB)和转化生长因子-β1(transforming growth factor-β1,TGF-β1)mRNA表达变化;采用ELISA方法检测IL-1β、IL-2、IL-6、TNF-α、IFN-γ蛋白含量变化;采用Western-blot检测NF-κB和TGF-β1蛋白含量变化;采用冰冻切片、HE染色和普鲁士蓝染色法检测脾脏组织病理学变化及含铁血黄素分布情况。分析铁过量后绵羊脾脏主要炎症因子的表达及病理形态学改变。结果发现,铁过量后绵羊脾脏出现含铁血黄素沉积,组织损伤严重。同时,脾脏IL-2、IL-6、NF-κB和TGF-β1 m RNA表达及蛋白含量显著升高,而IL-1β、TNF-α和IFN-γm RNA表达及蛋白含量均降低,且随着铁剂量的增加而呈对应关系。In order to explore the effect of excessive iron on main inflammatory factors and histopathology of sheep spleen,sixteen Merino hybrid rams were randomly divided into 4 groups:control group(CON),iron excess group 1(IronE1),iron excess group 2(IronE2)and iron excess group 3(IronE3).In 4 groups,ferrous sulfate monohydrate(FeSO4·H2 O)was added to basal diet with a dosage of 50,500,1000 and 1500 mg/kg,respectively.Sheep were slaughtered and spleens were taken out after feeding for 75 days.The m RNA expression changes of related inflammatory factors in the spleen of sheep were detected by fluorescence quantitative PCR technology,including interleukin-1β(IL-1β),interleukin-2(IL-2),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),interferon-γ(IFN-γ),nuclear factor-κB(NF-κB)and transforming growth factor-β1(TGF-β1).Changes of protein content were detected by ELISA,including IL-1β,IL-2,IL-6,TNF-α,IFN-γ,NF-κB and TGF-β1 were detected by western-blot.And histopathological changes and distribution of hemosiderin were detected by frozen sections and HE staining and prussian blue staining.Severe tissue damage and hemosiderin deposits were found in the spleen of sheep with excess iron.And the m RNA expression and pro tein content of IL-2,IL-6,NF-κB and TGF-β1 in the spleen increased significantly,while the m RNA expression and protein content of IL-1β,TNF-αand IFN-γdecreased,accompanied by a corresponding relationship with the increase in dose.
