摘  要:目的探索冠脉微血管三维可视化方法,观察双参宁心胶囊对大鼠冠状动脉微循环障碍(coronary microvascular dysfunction,CMD)冠脉微血管三维形态是否有改善。方法雄性健康SD大鼠随机分为三组:假手术组、模型组和双参宁心胶囊组,每组6只。双参宁心胶囊(90 mg/kg)组动物连续7 d灌胃给药剂量为1 mL/100 g,假手术和模型组灌胃等体积生理盐水,7 d后建立CMD模型,术后24 h处死取材。进行冠脉血管铸型及图像采集;开展心脏组织X射线衍射增强成像,将获得断层图像进行三维重建,观察冠脉微血管分布情况。结果使用自凝牙托材料可以快捷的制作大鼠冠脉血管铸型标本,可分辨的最小血管直径约为40μm;X射线衍射增强图像三维重建后得到冠脉微血管网的三维模型,最小可分辨直径10μm血管。血管铸型和X射线衍射增强成像均在一定程度上反应大鼠CMD时冠脉微血管空间分布改变,且可以观察到双参宁心胶囊干预对动物模型的冠脉微血管明显改善。结论X射线衍射增强成像方法能够在一定程度上构建出冠脉微血管网的三维可视化模型,可以观察到CMD大鼠冠脉微血管异常情况以及双参宁心胶囊具有增加冠脉微血管密度的作用。Objective To explore the three-dimensional visualization method of coronary microvessels,and whether Shuangshen Ningxin capsule improves the three-dimensional morphology of coronary microvessels in rats with coronary microvascular dysfunction(CMD).Method Male healthy SD rats were randomly divided into 3 groups,the sham operation group,the model group and the Shuangshen Ningxin capsule group.Shuangshen Ningxin capsule(90 mg/kg)group animals were given intragastric administration for 7 consecutive days.The sham operation group and the model group were given an equal volume of normal saline gavage.Seven days later,the coronary microcirculation dysfunction model was established,and the specimens were sacrificed 24 hours after the operation.Coronary vascular casting and image acquisition was conducted.The X-ray diffraction enhanced images were obtained,the tomographic images were used for the three-dimensional reconstruction,3 D model was built to observe the distribution of coronary microvessels.Result Using self-curing dental tray materials can quickly make rat coronary vascular cast specimens,the smallest resolvable blood vessel diameter is about 40μm.A three-dimensional model of the coronary microvascular network is obtained after the three-dimensional reconstruction of the X-ray diffraction enhanced image,the smallest resolvable blood vessel can reach 10μm.Both vascular casting and X-ray diffraction enhanced imaging reflect the changes in the spatial distribution of coronary microvessels in CMD rats,also the intervention of Shuangshen Ningxin capsule can also significantly improve the coronary microvasculature of animal models.Conclusion The X-ray diffraction enhanced imaging method can construct a three-dimensional visualization model of the coronary microvascular network to a certain extent,can reflect the abnormality of coronary microvascular in rats with coronary microvascular dysfunction,the effect of Shuangshen Ningxin capsule in increasing coronary microvessel density can also be reflected by this method
