摘 要:目的分析2018—2019年北京市朝阳区腮腺炎病毒(mumps virus, MuV)小疏水蛋白(small hydrophobic, SH)编码基因序列变异特点, 掌握朝阳区MuV型别构成及其变化规律, 为朝阳区流行性腮腺炎(简称流腮)防控提供基础资料。方法对2018—2019年朝阳区临床诊断为流腮患者的腮腺管口拭子样本进行MuV核酸检测及病毒分离培养, 并对MuV进行SH段基因扩增、序列分析和基因分型。结果 362份流腮患者的腮腺管口拭子样本中检出30份MuV核酸阳性标本, 并从中分离出16株MuV毒株, 将其SH基因序列与各基因型参考株和疫苗株序列进行比对, 其中15株MuV毒株为F基因型, 其核苷酸一致性为95.98%~100.00%, 氨基酸同源性为91.23%~100.00%, F基因型MuV毒株(19009株)的第28~30位置上的氨基酸三联体为IIL;1株MuV毒株为G基因型, 其与辽宁流行的G基因型毒株核苷酸一致性为98.85%, 氨基酸同源性为98.25%。本研究中F基因型与G基因型MuV毒株的核苷酸差异为12.07%。16株MuV北京分离株与Jeryl-Lynn疫苗株的核苷酸差异为13.22%~18.39%。结论 2018—2019年北京市朝阳区流腮病例中检出的MuV存在F和G两种基因型, 并以F基因型流行为主, 建议进一步加强对流腮的病原学监控。Objective To analyze the sequence variation characteristics of small hydrophobic(SH)gene of mumps virus(MuV)strains in Chaoyang district of Beijing from 2018 to 2019,so as to understand the composition and variation of MuV genotypes in this region,and provide basic data for mumps prevention and control in Chaoyang district.Methods MuV nucleic acid detection and cell culture were carried out for the swab samples at orifice of parotid duct collected from clinically diagnosed epidemic mumps cases in the sentinel hospitals of Chaoyang district from 2018 to 2019 and PCR amplification,sequencing analysis and phylogenetic analysis of SH segment of MuV strains were carried out.Results From 362 swab samples at orifice of parotid duct of epidemic mumps cases,30 were positive for MuV nucleic acid,and 16 MuV strains were isolated.The SH gene sequences of the strains were compared with the reference strains and vaccine strains of each genotype.The 15 MuV strains belonged to genotype F.The homology of nucleotide and amino acid sequences of the strains were 95.98%-100%and 91.23%-100%,respectively.The amino acids triplet at position 28-30 of the MuV strains of genotype F(strain 19009)was IIL.One strain belonged to genotype G.The homology of nucleotide and amino acid sequences between the strain and the strains of G genotype dominant in Liaoning were 98.85%and 98.25%,respectively.The differences in nucleotide sequences between strains of MuV genotype F and G in this study was 12.07%.The differences in nucleotide sequences between 16 strains of MuV isolated in Beijing and Jeryl-Lynn vaccine strain were 13.22%-18.39%.Conclusions Both MuV Genotype F and G were detected among epidemic mumps case in Chaoyang district of Beijing from 2018 to 2019 and genotype F was dominant.It is necessary to strengthen further the etiological monitoring of mumps.
