摘 要:本研究旨在从形态学和分子水平对牛羽虱(Bovicola bovis,B.bovis)进行鉴定。对采自湖南省长沙市黄牛体表的虱子样品进行初步形态学观察后,通过PCR对虫体线粒体细胞色素c氧化酶第I亚基(cytochrome c oxidase subunit 1,cox1)基因进行扩增,应用DNAMAN软件开展序列分析,并利用Mega 5.0软件最大似然法(maximum likelihood method)构建系统发育树。结果显示:3个虫体样本扩增片段长度与预期均一致,为389 bp,且其cox1序列与GenBank收录的牛羽虱(基因登录号MH001191)序列相似性为99.0%~99.5%;3个样本均与牛羽虱位于同一分支。本研究从形态学和分子水平证明采集样本为牛羽虱,进一步说明线粒体cox1基因可作为羽虱的分类鉴定遗传标记。For the purpose of identifying Bovicola bovis(B.bovis)from morphological and molecular level,after preliminary morphological observation for lice samples collected from yellow cattle in Changsha City of Hunan Province,the mitochondrial cytochrome c oxidase subunit 1(cox1)gene was amplifed by PCR.The sequence was analyzed by DNAMAN software,and a phylogenetic tree was constructed by maximum likelihood method in Mega 5.0 software.The results showed that the lengths of the three amplifed fragments were all consistent with the expected one,which was 389 bp,and the similarity between its cox1 sequence and that of B.bovis(accession No.MH001191)registered in GenBank was 99.0%to 99.5%;all the three samples were located in the same branch as B.bovis.In conclusion,the lice samples were confrmed to be B.bovis from morphological and molecular level,which further verifed that mitochondrial cox1 gene could be used as a genetic marker to classify and identify Bovicola.
