姜黄素诱导类风湿关节炎滑膜细胞凋亡及其机制研究

摘　　要：目的观察姜黄素(CUR)对体外培养的类风湿关节炎成纤维样滑膜细胞(RA-FLSs)凋亡的诱导作用及其机制。方法用不同浓度CUR处理在体外培养的RA-FLSs细胞后,CCK-8法检测CUR对细胞增殖能力的影响;流式细胞术检测CUR对RA-FLSs细胞凋亡的诱导效应;蛋白质免疫印迹方法(WB)检测细胞凋亡的外源途径中相关蛋白Fas,Caspase-3,Caspase-8的表达水平。结果CUR浓度为20、40、60μmol/L时可以显著抑制RA-FLSs的细胞的生长和增殖能力(P<0.05);流式检测结果表明,与对照组相比,CUR处理组早期凋亡细胞比例与晚期凋亡比例明显升高;WB检测结果显示,与对照组相比,CUR处理后,凋亡相关蛋白Fas、Caspase-3与Caspase-8表达显著增高(P<0.05)。结论CUR可以通过调节Fas凋亡通路,诱导RA-FLSs细胞发生凋亡。Objective To observe the effect of curcumin(CUR)on the apoptosis of rheumatoid arthritis fibroblast-like synoviocytes(RA-FLSs)cultured and its mechanism.Methods After treating RA-FLSs cells cultured with different concentrations of CUR,the effect on proliferation and apoptosis of RA-FLSs cells was detected using CCK-8 assay,flow cytometry,Western blot(WB),respectively.The relative expression of Fas,Caspase-3,Caspase-8 were detected by WB.Results CUR has a significant inhibitory effect on the growth and proliferation of RA-FLSs cells(P<0.05),and presents a dose-dependent effect;therefore,based on this result,the follow-up drug concentration is determined;follow-up experimental results show that different concentrations of CUR significantly induced apoptosis(P<0.05);the results of WB analysis showed that after CUR treatment,the expression of apoptotic proteins Fas,Caspase-3 and Caspase-8 increased significantly(P<0.05).We can draw a conclusion that CUR inhibits the proliferation of RA-FLSs by inducing the apoptosis.Conclusion CUR can induce apoptosis of RA-FLSs by regulating the Fas apoptotic signaling pathway.
