摘 要:目的:探讨18β-甘草次酸(18β-GA)通过下调钙黏蛋白相关蛋白(CTNNB1)表达对肺腺癌A549细胞放射敏感性的影响。方法:采用不同浓度的18β-GA(0,20,40,80μg·ml^(-1))处理肺腺癌A549细胞48 h,分别命名为对照组、18β-GA-20组、18β-GA-40组、18β-GA-80组;对肺腺癌A549细胞进行转染,分为CTNNB1小干扰RNA(si-CTNNB1)组、si-CTNNB1阴性对照(si-NC)组、CTNNB1过表达物(OE-CTNNB1)组、OE-CTNNB1阴性对照(OE-NC)组,转染24 h后,在OE-NC组、OE-CTNNB1组中分别加入40μg·ml^(-1)18β-GA处理48 h,并分别命名为18β-GA-40+OE-NC组、18β-GA-40+OE-CTNNB1组;测定肺腺癌A549细胞增殖、凋亡、CTNNB1 mRNA和蛋白表达水平以及肺腺癌A549细胞放射敏感性。结果:与对照组比较,18β-GA-20组、18β-GA-40组、18β-GA-80组肺腺癌A549细胞增殖抑制率、细胞凋亡率显著升高,细胞存活分数、CTNNB1 mRNA及蛋白表达水平显著降低,且呈浓度依赖性(P<0.05);18β-GA-20组、18β-GA-40组、18β-GA-80组放射增敏比(SER)值分别为1.476,1.736,2.392。与si-NC组比较,si-CTNNB1组肺腺癌A549细胞增殖抑制率、凋亡率显著升高,CTNNB1蛋白表达水平及细胞存活分数显著降低(P<0.05);si-CTNNB1组SER值为1.873。与18β-GA-40+OE-NC组比较,18β-GA-40+OE-CTNNB1组肺腺癌A549细胞增殖抑制率、凋亡率显著降低,CTNNB1蛋白表达水平及细胞存活分数显著升高(P<0.05);18β-GA-40组与18β-GA-40+OE-CTNNB1组SER值分别为1.736,0.592。结论:18β-GA可能通过下调CTNNB1表达来抑制细胞增殖、促进细胞凋亡,进而增强放射敏感性。Objective:To investigate the effects of 18β-glycyrrhetinic acid(18β-GA)on the radiosensitivity of lung adenocarcinoma A549 cells by down-regulating the expression of cadherin-associated protein beta 1(CTNNB1).Methods:Different concentrations of 18β-GA(0,20,40,80μg·ml^(-1))were used to treat lung adenocarcinoma A549 cells for 48 h,and the cells were named as control group,18β-GA-20 group,18β-GA-40 group and 18β-GA-80 group.Lung adenocarcinoma A549 cells were transfected and divided into CTNNB1 small interfering RNA(si-CTNNB1)group,si-CTNNB1 negative control(si-NC)group,CTNNB1 overexpression(OE-CTNNB1)group and OE-CTNNB1 negative control(OE-NC)group.After 24 hours of transfection,40μg·ml^(-1)18β-GA was added to the OE-NC group and OE-CTNNB1 group for 48 hours,and named as 18β-GA-40+OE-NC group and 18β-GA-40+OE-CTNNB1 group.The proliferation,apoptosis,CTNNB1 mRNA and protein expression levels and radiosensitivity of lung adenocarcinoma A549 cells were measured.Results:Compared with those in the control group,the proliferation inhibition rate and apoptosis rate of lung adenocarcinoma A549 cells in 18β-GA-20 group,18β-GA-40 group and 18β-GA-80 group were significantly increased,while the cell survival score,CTNNB1 mRNA and protein expression levels were significantly reduced,and the effects were concentration-dependent(P<0.05);the sensitization enhancement ratio(SER)values of 18β-GA-20 group,18β-GA-40 group and 18β-GA-80 group were 1.476,1.736 and 2.392,respectively.Compared with those in si-NC group,the proliferation inhibition rate and apoptosis rate of lung adenocarcinoma A549 cells in si-CTNNB1 group were significantly increased,and the CTNNB1 protein expression levels and cell survival score were significantly reduced(P<0.05);the SER value of si-CTNNB1 group was 1.873.Compared with those in 18β-GA-40+OE-NC group,the proliferation inhibition rate and apoptosis rate of lung adenocarcinoma A549 cells in 18β-GA-40+OE-CTNNB1 group were significantly reduced,and the CTNNB1 protein expression levels
