摘 要:目的:考察二氧化硅纳米粒(MS)不同的介孔结构对吸附西罗莫司自微乳(SRL-SMEDDS)性能的影响。方法:制备3 nm孔径的MSN-1和6 nm孔径的MSN-2。MS表面具有丰富的硅羟基(MS-H),对MS-H进行甲基(MS-M)、氨基(MS-A)和羧基(MS-C)的后嫁接修饰。使用扫描电镜、透射电镜、氮气吸附-脱附性能、动态光散射、水分散性和红外光谱等表征MS的不同介孔结构;通过比较粉体学参数、比表面积和孔容的变化,考察SRL-SMEDDS与MS的吸附比例;通过含量均匀度和重分散试验,评价固化制剂[SRL-SMEDDS-MS(-R)]的理化性质。结果:不同孔径的MS粉体学性能无明显差别,MSN-2吸附能力略大于MSN-1。经过表面官能团修饰,MS-M粉体学参数变化最小,比表面积与孔容的损失最小,保持较好的吸附性能。结论:MS的孔径和表面属性均对吸附性能有一定影响,其中,表面官能是影响MS吸附SRL-SMEDDS的关键因素。Objective:To investigate the effects of slilica nanoparticles(MS)with different mesoporous structures on the adsorption performance of sirolimus self-microemulsifying drug delivery system(SRL-SMEDDS).Methods:MSN-1 with 3 nm pore size and MSN-2 with 6 nm pore size were prepared.The surface of MSN had rich silanol groups(MS-H),and MS was respectively modified with methyl(MS-M),amino(MS-A)and carboxyl groups(MS-C).Different mesoporous structures of MS were characterized by scanning electron microscopy,transmission electron microscopy,N;adsorption-desorption,dynamic light scattering,water dispersibility and infrared spectroscopy.The adsorption ratio of SRL-SMEDDS and MS was studied by the powder parameters and the changes of specific surface area and pore volume.The physical and chemical properties of solidified formulations were evaluated by content uniformity and redispersion experiments.Results:There was no significant difference in powder performance of MS with different pore sizes.The adsorption capacity of MSN-2 was slightly larger than that of MSN-1.After surface functional group modification,MS-M powder had the smallest changes in the powder parameters,the loss of specific surface area and pore volume was minimal,and maintained good adsorption performance.Conclusion:Both the pore size and surface properties of MS have certain effects on the adsorption performance,and the surface functional group is the key factor affecting the adsorption of SMEDDS by MS.
