摘  要:目的探究负载鸢尾素微溶胶的静电纺丝支架对神经干细胞分化的影响。方法用微溶胶静电纺丝技术制备负载鸢尾素的静电纺丝支架(PLLA),使用透射电镜(TEM)检测纤维支架的内部结构,使用扫描电镜(SEM)检测纤维支架的形貌特征。通过等离子技术将鸢尾素枝接于静电纺丝支架,并测定其释放曲线。提取SD大鼠胎鼠海马区神经干细胞,通过免疫荧光染色进行细胞标志物巢蛋白(Nestin)的鉴定。观察PLLA-鸢尾素浸出液对神经干细胞分化的影响。结果TEM显示静电纺丝支架有管状结构,SEM显示其有一致的方向特征;鸢尾素可持续释放48h;提取的细胞聚团呈现出明显的球形,免疫荧光染色表明其表达NeStin;PLLA-鸢尾素浸出液可以促进神经干细胞分化。结论负载鸢尾素微溶胶静电纺丝支架可以促进神经干细胞的分化。Objective To investigate the effect of electrospinning scaffold loaded with irisin microsol on the differentiation of neural stem cells.Methods The electrospinning scaffolds loaded with irisin(PLLA)were prepared by microsol electrospinning technology.The internal structure of the fiber scaffold was examined by transmission electron microscopy(TEM)and the morphological characteristics of the fiber scaffold were detected by scanning electron microscope(SEM).Irisin was attached to electrospinning scaffolds by plasma technology,and its release curve was measured.Neural stem cells from the hippocampus of SD rat fetus were extracted and identified by immunofluorescence staining.The effect of PLLA-irisin extract on differentiation of neural stem cells was observed.Results TEM showed that the electrospinning scaffold had tubular structure,and SEM showed that it had consistent directional characteristics.The irisin could be released continuously for 48 hours.The extracted cell clusters showed obvious spherical shape,and immunofluorescence staining showed that they expressed Nestin.The PLLA-irisin extract can promote the differentiation of neural stem cells.Conclusion The microsol electrospinning scaffolds loaded with irisin can promote the differentiation of neural stem cells.
