摘 要:目的:探究MACF1在小鼠牙乳头细胞向成牙本质细胞向分化中的作用。方法:免疫组化检测MACF1在不同发育时期小鼠磨牙胚中的表达情况。建立小鼠牙乳头细胞向成牙本质细胞向分化的模型,使用RT-qPCR和Western Blot检测MACF1在此过程的表达。使用MACF siRNA抑制MACF1的表达,检测其对小鼠牙乳头细胞向成牙本质细胞向分化的影响。结果:MACF1在胚胎期16.5 d(E16.5)和出生后1 d(PN1)的小鼠磨牙胚中的牙乳头间充质细胞中有微弱表达,在成牙本质细胞层中大量表达。体外实验显示在小鼠牙乳头细胞向成牙本质细胞向分化过程中,MACF1的表达上升。抑制细胞内MACF1的表达后,细胞内成牙本质细胞向分化的相关基因表达减少。结论:MACF1促进小鼠牙乳头细胞向成牙本质细胞向分化。Objective:To investigate the function of MACF1 during the odontoblastic differentiation of mouse dental papilla cells.Methods:Immunohistochemistry was used to detect the expression of MACF1 in the mouse molars at different development stage.After establishing the model of odontoblastic differentiation of mouse dental papilla cells, RT-qPCR and Western blot were employed to the detect the expression of MACF1 in this process.Western blot was used to examine the effect of MACF1 on odontoblastic differentiation of mouse dental papilla cells after transfecting with MACF1 siRNA.Results:MACF1 shows a moderate level in dental mesenchymal cells in the mouse molar molars at E16.5 and PN1,and it is highly expressed in the odontoblasts at PN1.The expression of MACF1 increased during the odontoblastic differentiation of mouse dental papilla cells in vitro.After inhibiting MACF1,the expression of related genes of odontoblastic differentiation decreased.Conclusion:MACF1 promotes the odontoblastic differentiation of mouse dental papilla cells.
