摘  要:目的实验旨在探讨电针对动脉粥样硬化家兔肠道菌群和5-羟色胺(5-HT)的影响。方法选择新西兰白兔27只,随机分为5组:对照组(n=5)、模型组(n=5)、阿托伐他汀组(n=5)、电针组(n=5)和电针预处理组(n=5),剩下2只造模后处死,观察其颈总动脉病理变化以确认建模成功。除对照组外,其余各组均采用高脂饮食,并施以颈动脉球囊损伤术,建立炎症介导的血管内皮损伤动脉粥样硬化模型。造模后,电针组和阿托伐他汀组分别进行电针处理和服用阿托伐他汀钙片,每天1次,每周6 d,持续1个月。电针预处理组,在建模前1个月予电针治疗(针刺方法与电针组相同),造模后再施以与电针组相同的电针处理。观察门水平肠道菌群及血清5-HT水平变化。结果与模型组比较,电针预处理组、电针组和阿托伐他汀组,拟杆菌门丰度和中枢5-HT含量明显升高(P<0.05或P<0.01),厚壁菌门丰度和血清5-HT含量明显降低(P<0.05或P<0.01)。其中,电针预处理组对于厚壁菌和拟杆菌的改变更为显著。结论电针可抑制动脉粥样硬化(AS)发展的内在机制,可能与影响肠道菌群拟杆菌门丰度、厚壁菌门丰度、血清和中枢5-HT含量有关。Objective This study was to explore the effects of electro-acupuncture(EA)on the reconstruction of intestinal microflora and the influence on the 5-HT in the model of atherosclerotic rabbit intestine,as well as its function. Methods A total of 27 New Zealand white rabbits were selected and randomly divided into 5 groups:the control group(n=5),the model group(n=5),the atorvastatin group(n=5),the EA group(n=5)and the EA pretreatment group(n=5),the other 2 rabbits were sacrificed to confirm the success of the modeling. Except for the control group,the rabbits of all groups were subjected to high-fat diet and then combined it with carotid artery balloon injury to establish the atherosclerotic model of inflammation-mediated vascular endothelial injury. Subsequently,the atorvastatin group was given atorvastatin calcium tablets,once a day,6 days a week for a month;and an EA treatment was given to the EA group,once a day,6 days a week for a month. For the EA pretreatment group,a month of EA prevention was applied before the modeling;after modeling the same EA treatment as EA group was applied. The changes of intestinal flora and serum 5-HT were observed. Results For the EA pretreatment group,the EA group and the atorvastatin group,the results showed that all the three treatment methods can increase the number of Firmicutes and the content of brain 5-HT(P<0.05 or P<0.01),decrease the number of Bacteroides and the content of serum 5-HT(P<0.05 or P<0.01). While for the results of the number of Firmicutes/Bacteroidetes and the regulating of abundance of intestinal flora,the EA pretreatment group showed better efficacy compare to EA group and atorvastatin group. Conclusion The mechanism of EA inhibiting the development of as may be related to the effect of EA on the contents of serum 5-HT and brain 5-HT and the intestinal number of Bacteroides and Firmicutes.
