摘 要:目的通过建立乙醇-三硝基苯磺酸(TNBS)诱导溃疡性结肠炎模型,研究蒲公英水提物对溃疡性结肠炎大鼠的治疗作用,并初步探索其作用机制。方法采用三硝基苯磺酸诱导溃疡性结肠炎大鼠模型,灌肠给予奥沙拉嗪(200 mg/kg)和蒲公英水提物(100、200 mg/kg),通过检测大鼠疾病活动指数(DAI)、结肠黏膜损伤指数(CMDI)和结肠病理评分(HS)以及结肠长度、宽度,酶联免疫吸附(ELISA)法检测血清C反应蛋白(CRP)、白细胞介素-8(IL-8)、IL-10、IL-17、肿瘤坏死因子-α(TNF-α)及结肠髓过氧化物酶(MPO)、IL-2、表皮生长因子(EGF)、转化生长因子-β(TGF-β)来评价蒲公英水提物对溃疡性结肠炎大鼠的作用。结果与模型组比较,蒲公英水提物组的DAI、CMDI、HS评分均降低(P<0.05,P<0.01),胸腺指数升高(P<0.05,P<0.01),脾脏指数和结肠指数降低(P<0.01),结肠长度和结肠长宽比增加(P<0.05,P<0.01)。同时,蒲公英水提物能降低CRP、MPO活性(P<0.01),抑制促炎细胞因子IL-2、IL-8、IL-17、TNF-α水平(P<0.01),升高抗炎细胞因子IL-10和促修复因子EGF水平(P<0.01)。结论蒲公英水提物对TNBS诱导的溃疡性结肠炎大鼠有治疗作用,其机制可能是通过调节炎性细胞因子分泌,减轻机体炎症反应,提高促修复因子表达,促进黏膜愈合,改善结肠黏膜完整性有关。AIM To study the therapeutic effects of aqueous extract of dandelion on TNBS/ethanol-induced ulcerative colitis in rats, and initially explore its mechanism of action. METHODS Upon ulcerative colitis rat models induced by TNBS/ethanol, enema of olsalazine(200 mg/kg) or aqueous extract of dandelion(100, 200 mg/kg) was administered. Subsequently, the rats were subjected to the measurement of DAI, CMDI and HS scores and length and width of ulcerative colitis;the detection of serum levels of CRP, IL-8, IL-10, IL-17, TNF-α and colonic levels of MPO, IL-2, EGF, TGF-β by ELISA. RESULTS Compared with the model group, the groups intervened with aqueous extract of dandelion shared decrease in scores of DAI, CMDI and HS(P<0.05, P<0.01) and levels of spleen index and colon index(P<0.01), increase in thymus index level(P<0.05, P<0.01) and length and aspect ratio of the colon(P<0.05, P<0.01). Simultaneously, reduced activities of CRP and MPO(P<0.01), inhibited levels of pro-inflammatory cytokines IL-2, IL-8, IL-17 and TNF-α(P<0.01), and increased levels of anti-inflammatory cytokine IL-10 and the pro-repair factor EGF were noticed after the treatment with aqueous extract of dandelion(P<0.01). CONCLUSION Aqueous extract of dandelion has a therapeutic effect on TNBS/ethanol-induced ulcerative colitis rats. The mechanism may be related to its efficacy in regulating the secretion of inflammatory cytokines, reducing the inflammatory response, increasing the expression of pro-repair factors, thereby promoting mucosal healing, and improving colonic mucosal integrity.
