摘　　要：为了获得低成本高产量的抗原表达,开发新型、有效防控猪圆环病毒的亚单位疫苗,根据猪圆环病毒2型(PCV2)Cap蛋白序列,通过软件设计优化了密码子并合成cap基因,利用多角体病毒作为载体,构建了表达PCV2 Cap蛋白的重组病毒,命名为flashBAC-PCV2-ORF2株。经过鉴定,证明该病毒株表达Cap蛋白的抗原量在150μg/mL以上,毒株纯净,没有细菌、支原体和其他病毒污染。经纯化后,通过猪免疫试验证明,构建的flashBAC-PCV2-ORF2株具有良好免疫原性,能够对病毒攻击起到很好的免疫保护作用,具有较高的潜在应用价值。Based on the Cap protein of porcine circovirus type 2(PCV2),apotential vaccine strain with high expression of Cap protein was constructed.According to the published PCV2 Cap protein sequence,the codon was optimized by software design and cap gene was synthesized.Using polyhedrosis virus as vector,the recombinant virus expressing PCV2Cap protein was constructed,named flashBAC-PCV2-ORF2strain.After identification,it was proved that the antigen quantity of Cap protein expressed by the virus strain was over 150μg/mL,the virus strain is pure,no bacteria,mycoplasma and other virus pollution.The purified Cap protein was proved to have good immunogenicity and protection against virus challenge by pig immune test.The results showed that the constructed viral vector and Cap protein expressed have good immunogenicity,and potential application value of vaccine.
