摘  要:目的 落新妇苷是广泛存在于多种植物中的二氢黄酮醇苷,黄酮类化合物的有效成分,具有抗炎、抗氧化、抗病毒等多种药理活性。文章主要探讨落新妇苷对oxLDL诱导的血管平滑肌细胞凋亡、炎症反应的影响及分子机制。方法 人主动脉血管平滑肌细胞(HA-VSMC)随机分为对照组、oxLDL组、oxLDL+落新妇苷低、中、高剂量组、oxLDL+miR-NC组、oxLDL+miR-613组、oxLDL+落新妇苷+anti-miR-NC组、oxLDL+落新妇苷+anti-miR-613组;流式细胞术检测细胞凋亡;Western blot法检测蛋白表达;酶联免疫吸附试验(ELISA)检测IL-1β、TNF-α水平;实时荧光定量PCR(RT-qPCR)检测miR-613表达水平。结果 不同剂量落新妇苷处理后,oxLDL诱导的血管平滑肌细胞的凋亡率降低,cleaved-caspase3和cleaved-caspase9蛋白表达水平降低,IL-1β、TNF-α水平降低,miR-613表达水平升高,呈剂量依赖性(P<0.05)。过表达miR-613后,血管平滑肌细胞的凋亡率降低,cleaved-caspase3和cleaved-caspase9蛋白表达水平降低(P<0.05)。与oxLDL+miR-NC组IL-1β水平[(43.33±4.23)pg/mL]、TNF-α水平[(89.15±5.61)pg/mL]比较,oxLDL+miR-613组[(17.98±1.26)、(46.31±4.59)pg/mL]降低(P<0.05)。结论 落新妇苷通过上调miR-613的表达抑制oxLDL诱导的血管平滑肌细胞凋亡、炎症反应。Objective Astilbin is a dihydroflavanol glycoside widely existing in a variety of plants, and meanwhile the effective component of flavonoids includes anti-inflammatory, antioxidant, antiviral and other pharmacological activities. In this paper, the effects and molecular mechanism of astilbin on apoptosis and inflammation of vascular smooth muscle cells induced by oxLDL were investigated. Methods Human aortic vascular smooth muscle cells(HA VSMC) were randomly divided into control group, oxLDL group, oxLDL+ astilbin low, medium and high dose groups, oxLDL+ miR-NC group, oxLDL+ miR-613 group, oxLDL+ aspartine + anti-miR-NC group, oxLDL+ astilbin + anti miR-613 group;Cell apoptosis was detected by flow cytometry. Western blot was used to detect protein expression. Enzyme-linked immunosorbent assay(ELISA) was used to detect Il-1 β and TNFα levels. The expression level of miR-613 was detected by real-time fluorescence quantitative PCR(RT-QPCR). Results After treatment with different doses of astilbin, the apoptosis rate of vascular smooth muscle cells induced by oxLDL was decreased, cleaved caspase3 and caspase9 protein expression levels were decreased, IL-1β and TNFα levels were decreased, and miR-613 expression level was increased in a dose-dependent manner(P<0.05). After overexpression of miR-613, the apoptosis rate of vascular smooth muscle cells was decreased, and cleaved caspase3 and caspase9 protein expression levels were decreased(P<0.05). Compared with the levels of IL-1β [(43.33±4.23) pg/mL] and TNFα [(89.15±5.61) pg/mL] in oxLDL+miR NC group, OxLDL + miR-613 group [(17.98±1.26),(46.31±4.59) pg/mL] decreased(P<0.05). Conclusion Astilbin can inhibit the apoptosis and inflammation response of vascular smooth muscle cells induced by oxLDL by the up-regulation of miR-613 expression.
