摘  要:为探究猪圆环病毒2型(PCV2)Cap蛋白与热休克蛋白DNAJC7的关系,用Cap与DNAJC7蛋白的重组杆状病毒质粒pFast-GST-Cap和pFast-DNAJC7共转染sf9细胞,并用免疫共沉淀试验(Co-IP)检测。结果显示,沉淀Cap蛋白后能检测到DNAJC7蛋白条带,且沉淀DNAJC7蛋白后能够检测到Cap蛋白条带,表明Cap蛋白与DNAJC7蛋白之间存在相互作用。论文完善了PCV2 Cap蛋白参与的复杂分子间相互作用网络,并为阐明DNAJC7蛋白对PCV2复制的影响及机制奠定了基础。To explore the interaction between porcine circovirus type 2(PCV2)Cap protein and heat shock protein DNAJC7,the recombinant baculovirus plasmids pFast-GST-Cap and pFast-DNAJC7 were co-transfected into SF9 cells and detected by co-immunoprecipitation assay(Co-IP).The results showed that the protein band of DNAJC7 could be detected after the precipitation of Cap protein,and the protein band of Cap could be detected after the precipitation of DNAJC7 protein,indicating that there is an interaction between Cap protein and DNAJC7 protein.This study further improved the complex intermolecular interaction network involved in the PCV2 Cap protein,and laid a foundation for the clarification of the effect of DNAJC7 protein on PCV2 replication and its mechanism.
