摘 要:对2017年-2018年采自中国集中生猪养殖区的疑似猪繁殖与呼吸综合征病毒(PRRSV)感染的309份猪肺脏组织样品的ORF5基因序列进行测定和遗传进化分析,结果表明,PRRSV阳性数119份,占检测样品数的38.51%,其中HP-PRRSV毒株91份,占总阳性样品数的76.5%;类NADC30毒株28份,占阳性样品总数的23.5%。阳性样品中有53株GP5的氨基酸相似性为87.1%~100%,与北美型毒株VR-2332氨基酸的相似性为79.9%~96.6%,与HP-PRRSV代表株JXA1的氨基酸相似性在79.9%~96.6%,与类NADC30代表株HENAN-XINX的氨基酸相似性为80.5%~93.3%。多序列在诱骗表位(27-30aa)存在氨基酸突变,由A^(29)突变为V^(29)。主要中和表位PNE(37-45aa)处也存在氨基酸突变,HP-PRRSV毒株L^(39)全部突变为I^(39),2018HZAU-FJNP-419等多毒株的PNE处L^(39)全部突变为S^(39)、H^(38)全部突变为Y^(38)。说明PRRSV在2017年-2018年仍在我国广泛流行并不断变异。In order to understand the prevalence and genetic variation of porcine reproductive and respiratory syndrome virus(PRRSV),309 pig tissue and serum samples suspected of PRRSV infection collected in major pig breeding areas in China during 2017-2018 were tested,and ORF5 gene sequencing and genetic evolution analysis were performed on PRRSV positive samples.The positive rate of PRRSV was 38.51%.Among them,there were 91 HP-PRRSV strains,accounting for 76.5%of the total positive samples;there were 28 NADC30-like strains,accounting for 23.5%of the total positive samples.The amino acid similarity of 53 strains of GP5 was between 87.1%-100%,79.9%-96.6%with the North American strain VR-2332,79.9%-96.6%with the representative strain JXA1 of HP-PRRSV,and 80.5%-93.3%with the representative strain HENAN-XINX of NADC30.There was an amino acid mutation in the decoy epitope(27-30aa)of multiple sequences,which changed from A29 to V29.There were also amino acid mutations at the main neutralization epitopes of PNE(37-45AA),all of the L39 strains of HP-PRRSV were mutated to I39,and all of the L39 strains of 2018HZAU-FJNP-419 were mutated to S39 and all of the H38 strains were mutated to Y38 at the PNE.The results showed that PRRSV was still widely prevalent and continuously mutated in China during 2017-2018.
