摘 要:目的:观察款冬花散对支气管扩张症痰热蕴肺证模型大鼠气道炎症的影响。方法:70只大鼠随机分为空白组、款冬花散自身对照组、模型组、罗红霉素组、款冬花散干预组(低、中、高剂量),每组10只。采用绿脓杆菌液气管注入制备支气管扩张症动物模型。采用款冬花散干预2周后取材,观察肺组织形态改变、肺泡灌洗液(BALF)中白三烯B4(LTB4)、肿瘤坏死因子-α(TNF-α)、白细胞介素-8(IL-8)、白细胞介素-10(IL-10)水平及肺组织LTB4、TNF-α、IL-8、IL-10 mRNA表达。结果:与空白组比较,模型组大鼠气道肺组织有大量炎性细胞浸润,BALF、肺组织的LTB4、TNF-α、IL-8、IL-10含量及mRNA表达增加(P<0.01)。款冬花散能降低气道肺组织炎症程度,减轻BALF中的LTB4、TNF-α、IL-8、IL-10水平,下调肺组织LTB4、TNF-α、IL-8、IL-10 mRNA表达,且以高剂量组较为明显(P<0.01)。结论:款冬花散在一定程度上降低支气管扩张症痰热蕴肺证模型大鼠气道炎症状态,并呈现一定的量效关系趋势。Objective:To observe the effect of Kuandonghua formula on airway inflammation in rats for bronchiectasis with the phlegm-heat stagnation syndrome.Methods:A total of 70 rats were randomly divided into the blank group,the self-control group,the model group,the roxithromycin group,the Kuandonghua formula treatment group at low,middle and high dosage with 10 rats in each group.The rat model was induced by intratracheal injection of pseudomonas aeruginosa dilution.Rats in the treatment group were treated with Kuandonghua formula for 2 weeks.The morphological changes of lung tissue,concentrations in bronchoalveolar lavage fluid(BALF)and expressions of mRNA in the lung tissue including LTB4,TNF-α,IL-8 and IL-10 were evaluated.Results:Compared with the blank group,the inflammatory infiltration of lung tissue,concentrations in BALF and expressions of mRNA in the lung tissue including LTB4,TNF-α,IL-8 and IL-10 of the model group were significantly increased(P<0.01).Compared with the model group,the inflammatory infiltration of lung tissue,concentrations in BALF and expressions of mRNA in the lung tissue with regard to LTB4,TNF-α,IL-8 and IL-10 of the Kuandonghua formula treatment group were significantly decreased,which the effect of high dosage was superior to low and middle dosage(P<0.01).Conclusion:Kuandonghua formula can reduce airway inflammation in rats for bronchiectasis with the phlegm-heat stagnation syndrome,and it presents a certain quantity-effect relationship trend.
