摘  要:樟树是我国亚热带地区重要的常绿阔叶树种,也是常见的园林绿化和特色经济树种。樟树根据枝叶精油主成分可划分为芳樟醇型、柠檬醛型、脑樟型等化学型,具有重要经济价值。以樟树赣芳1号枝条为材料,研究外植体采集时间、消毒时间、枝条木质化程度、生长调节剂等对樟树赣芳1号茎段组织培养的影响。结果表明,5月份采集1年生半木质化枝条,修剪成1~2芽茎段,用0.1%升汞消毒5 min,污染率和褐化率低,萌芽率高。污染率和褐化率随枝条木质化程度的增加而升高,萌芽率随枝条木质化程度的增加而降低。因此樟树赣芳1号茎段组织培养应采集1年生半木质化枝,萌芽率高、污染率和褐化率低。诱导茎段萌芽的最适培养基为MS+1.2 mg/L 6-BA+0.05 mg/L IBA,萌芽率为63.33%。最适增殖培养基为MS+1.0~2.0 mg/L 6-BA+0.1 mg/L IBA,增值系数为3.5。生根培养11 d组培苗出现不定根,最适生根培养基为1/2 MS+0.5 mg/L NAA,生根率达到93.33%。生根培养15~20 d,大棚炼苗7~10 d,移栽成活率可达90%以上。Cinnamomum camphora is one of the important species of evergreen broad-leaved forests in the subtropical regions of China,and it is also a common landscaping and characteristic economic tree species.According to the main components of essential oil in branches and leaves of C.camphora,it can be divided into several chemical types such as linalool,citral and camphor,which have high economic value.The effects of explant collection time,disinfection time,lignification degree and growth regulator on tissue culture of branches of C.camphora‘Ganfang 1’were studied.The results show that the axillary bud germination rate of stem segments from semi-lignified stem segments of C.camphora‘Ganfang 1’was the highest with 0.1%HgCl_(2) disinfected for 5 min in May,and the browning rate and pollution rate were low.The percentage of pollution and browning increased with the degree of lignification of branches,and the percentage of germination decreased with the degree of lignification of branches.Therefore,one-year-old semi-lignified shoots should be collected for stem segment tissue culture of C.camphora‘Ganfang 1’,with high germination rate,low pollution rate and browning rate.The most suitable medium for stem segment germination of C.camphora‘Ganfang 1’was MS+1.2 mg/L 6-BA+0.05 mg/L IBA,and the germination rate was 63.33%.The most suitable medium for multiplication was MS+1.0~2.0 mg/L 6-BA+0.1 mg/L IBA,the multiplication coefficient was 3.5.Adventitious roots begin to appear at the base of tissue culture seedlings for 11 days of rooting culture,and the most suitable medium for rooting was 1/2 MS+0.5 mg/L NAA,and the rooting rate reached 93.33%.After 15~20 days of rooting culture and 7~10 days of seedling training in plastic shed,the survival rate of transplanting could reach over 90%.
