摘  要:目的探讨妊娠特异性β1糖蛋白-1(PSG1)在重度子痫前期(SPE)孕妇胎盘组织及血清中的表达及其与SPE患者临床指标的相关性。方法收集2020年8月至2021年4月住院并分娩的孕妇共57例,其中重度子痫前期(SPE)21例、妊娠期高血压(GH)23例、正常对照组孕妇13例;收集其分娩前血清标本及分娩后的胎盘组织;应用real-time PCR及Western blot分别检测其胎盘组织中PSG1 mRNA及PSG1蛋白的表达;利用ELISA法检测血清PSG1水平,并分析血清PSG1水平与相关指标之间的相关性。结果随着病情的进展,无论在胎盘组织及血清中PSG1的表达均呈一定的下降趋势,具体表现为SPE孕妇胎盘组织及血清中PSG1的表达水平均显著低于GH及对照组孕妇(P<0.05);GH孕妇胎盘组织PSG1 mRNA水平低于对照组孕妇(P<0.05),而血清PSG1水平与对照组相比差异无统计学意义(P>0.05)。PSG1在SPE组孕妇血清中的水平与孕妇收缩压(r=-0.687,P<0.05)、24h尿蛋白定量(r=-0.781,P<0.05)呈显著负相关,与相应胎盘组织中PSG1 mRNA表达水平(r=0.673,P<0.05)呈正相关。结论PSG1在重度子痫前期孕妇胎盘组织及血清中的水平显著降低,提示与重度子痫前期的发生发展密切相关。Objective To explore the expression of pregnancy-specificβ1 glycoprotein-1(PSG1)in the placenta tissue and serum of pregnant women with severe preeclampsia(SPE)and its correlation with the onset of severe preeclampsia.Methods A total of 57 pregnant women who were hospitalized and gave birth from August,2020 to April,2021 were collected for the study,including 21 severe preeclampsia(SPE),23 hypertension during pregnancy(GH),and 13 pregnant women in the normal control group.The expression of PSG1 in placental tissue and serum were detected respectively,and the correlation between serum PSG1 level and clinical indicators was analyzed.Results The expression level of PSG1 in placenta tissue and serum of SPE pregnant women was significantly lower than that of GH and control pregnant women(P<0.05).The level of PSG1 mRNA in placenta of GH pregnant women was lower than that of the control group(P<0.05),while the level of serum PSG1 was not significantly different from that of the control group(P>0.05).The level of PSG1 in serum of pregnant women in the SPE group was significantly negatively correlated with the systolic blood pressure(r=-0.687,P<0.05)and 24h urine protein(r=-0.781,P<0.05),and was significantly negatively correlated with PSG1 mRNA in the corresponding placental tissues,while the expression level(r=0.673,P<0.05)was positively correlated.Conclusion As the disease progresses,the level of PSG1 in placental tissue and serum gradually increases,suggesting that it is closely related to the occurrence and development of preeclampsia.
