摘 要:为了探索百脉根根瘤内生菌多样性,揭示百脉根根瘤内根瘤菌与假单胞菌的多样性及其互作关系并寻找能结瘤的根瘤菌,本研究采用免培养和可培养相结合的方法对百脉根根瘤内生菌的多样性进行分析,采用平板对峙法对根瘤菌与假单胞菌的互作关系进行初步研究。结果表明:免培养法从百脉根根瘤内检测到内生菌分属于17门31纲69目103科140属,可培养法分离鉴定了3门4纲12目17科28属的内生细菌,根瘤菌与假单胞菌为优势种群。免培养法从百脉根根瘤内检测到根瘤菌有中慢生根瘤菌属、氨基杆菌属、茎根瘤菌属、新根瘤菌属、副根瘤菌属、根瘤菌属、剑菌属和申氏杆属,可培养法从百脉根根瘤内分离获得6属11种根瘤菌,分别为新泻氨基杆菌(TRM95017)、纤维素根瘤菌(TRM95031)、甘薯根瘤菌(TRM95046)、罗氏根瘤菌(TRM95060)、葡萄汁新根瘤菌(TRM95085)、附著剑菌(TRM95093)、鸡眼草申氏杆菌(TRM95114)、谷粒申氏杆菌(TRM95115)、动胶菌样申氏杆菌(TRM95116)、桑娟中慢生根瘤菌(TRM95624)和戈壁中慢生根瘤菌(TRM95240)。对可培养法分离获得的根瘤菌进行结瘤实验,从中筛选到一株结瘤根瘤菌为新泻氨基杆菌(TRM95017)。对峙法结果表明能够与根瘤菌融合共生的假单胞菌有鱼假单胞菌(TRM95006)、恶臭假单胞菌(TRM95007)、猴假单胞菌(TRM95024)等13种。研究表明百脉根根瘤内根瘤菌与假单胞菌物种多样性丰富,为生物固氮研究储备了重要的菌种资源。In order to explore the diversity of endophytic bacteria,reveal the interaction relationship between Rhizobia and Pseudo-monas within root nodules of Lotus japonicus,and find nodular rhizobia.The culture-dependent method and culture-independent method were choosed to analyze the diversity of endophytic bacteria in nodules of L.japonicus.The plate confrontation method was used to study the relationship between Pseudomonas and Rhizobium.The results showed that 140 genera,103 families,69 orders,31 classes and 17 phyla endophytic bacteria were detected from nodules by culture-independent method,and 28 genera,17 families,12 orders,4 classes and 3 phyla were obtained by culture-dependent method,the dominant genera were Rhizobium and Pseudomonas.By culture-independent method,the genera of rhizobia were mainly Mesorhizobium,Aminobacter,Allorhizobium,Neorhizobium,Pararhizobium,Rhizobium,Ensifer and Shinella.11 species,6 genara of Rhizobium were isolated from nodules of L.japonicus by culture-dependent method,which were Aminobacter niigataensis(TRM95017),Rhizobium cellulosilyticum(TRM95031),Rhizobium ipomoeae(TRM95046),Rhizobium rosettiformans(TRM95060),Neorhizobium vignae(TRM95085)Ensifer adhaerens(TRM95093),Shinella kummerowiae(TRM95114),Shinella granuli(TRM95115),Shinella zoogloeoides(TRM95116),Mesorhi-zobium sanjuanii(TRM95624),Mesorhizobium gobiense(TRM95240),respectively.A nodulation experiment resulted in obtaining a nodular rhizobia named as Aminobacter niigataensis(TRM95017).The plate confrontation method indicated that thirteen species of Pseudomonas were symbiotic with Rhizobium,such as Pseudomonas piscium(TRM95006),Pseudomonas putida(TRM95007)and Pseudomonas simiae(TRM95024).The research showed that Rhizobia and Pseudomonas are rich in the nodules of L.Japoni-cus,which was an important resources for nitrogen fixation research.
