摘  要:目的:研究玄参提取物对阴虚火旺甲状腺功能亢进(简称甲亢)大鼠基因表达谱的影响,探讨玄参提取物干预大鼠的分子机制。方法:采用转录组测序技术检测对照组、模型组和玄参组大鼠肝脏组织的基因表达谱,通过STEM软件筛选差异表达基因(DEGs),并针对DEGs进行GO功能富集KEGG通路分析。结果:与对照组比较,模型组筛选DEGs 79个;与模型组比较,玄参组筛选DEGs 5个。KEGG通路分析结果显示,Rap1、Hedgehog、Jak-STAT和PI3K-Akt信号通路,包含LPAR3、IHH和CNTFR,可能是玄参提取物治疗甲亢、发挥甲状腺保护作用的关键通路与基因。结论:玄参提取物干预阴虚火旺甲亢大鼠的分子机制与细胞生长、死亡相关基因和糖脂代谢相关基因密切相关,这为后续玄参治疗阴虚火旺甲亢的关键基因验证和作用机制研究提供了研究基础。Objective: To study the effects of extract from Radix Scrophulariae on gene expression profile of hyperthyroidism rat model of yin deficiency and fire excess, and to explore the molecular mechanism of the intervention. Methods:The gene expression profiles of control group, model group and Radix Scrophulariae group were detected by using transcriptome sequencing technique. The DEGs were screened by STEM trend analysis software. GO function enrichment and KEGG pathway analysis were performed for DEGs. Results: Compared with the control group, 79 DEGs were obtained in model group;compared with the model group, 5 DEGs were obtained due to Radix Scrophulariae group. The results of KEGG enrichment analysis indicated that Rap1, Hedgehog, Jak-STAT and PI3K-Akt pathway(LPAR3, IHH and CNTFR) may be the key pathways and genes in the treatment of hyperthyroidism and thyroid protection. Conclusion: The molecular mechanism of extract from Radix Scrophulariae in protection against hyperthyroidism rat model of yin deficiency fire excess is closely related to cell proliferation and apoptosis-related genes and glycolipid metabolism-related genes, which providing a research basis for the verification of key genes and the study of mechanism of Radix Scrophulariae in the treatment of yin deficiency and fire excess hyperactivity hyperthyroidism.
