摘  要:目的:观察复方铁皮石斛与厄贝沙坦和氨氯地平合用对自发性高血压大鼠(SHR)的降压疗效,并初步探讨其作用机制。方法:实验一:2月龄SHR大鼠随机分为模型组、复方铁皮石斛组(复方组)、厄贝沙坦组、厄贝沙坦+氨氯地平组(2西药组)、复方铁皮石斛+厄贝沙坦+氨氯地平组(2西药+复方组),每组10只,10只WKY大鼠为正常组,给药6周。实验二:8月龄SHR大鼠随机分为模型组、 2西药组、2西药+复方组,每组6只,5只WKY大鼠为正常组,给药4周。实验一和实验二大鼠灌胃后测定给药后3 h、24 h收缩压(SBP),实验二疗程结束后酶联免疫法检测血清血管紧张素Ⅱ(AngⅡ)及胰岛素含量,实时荧光定量PCR测定心脏AngⅡ的1型受体(AT1R)及2型受体(AT2R)、胰岛素受体(ISR)、胰岛素α受体(ISR-α)、胰岛素β受体(ISR-β)的基因表达。结果:在整个疗程的给药3 h、24 h后,与2西药组比较,2西药+复方组SBP均显著降低(P<0.01);与模型组比较,2西药+复方组的血清AngⅡ显著升高(P<0.05),2西药组、2西药+复方组AT1R mRNA表达显著升高(P<0.05);与2西药组比较,2西药+复方组的ISR mRNA表达显著升高(P<0.05)。结论:复方铁皮石斛与厄贝沙坦、氨氯地平合用降压增效显著,其机制可能与上调ISR mRNA表达有关。Objective: To explore the antihypertensive efficacy of Compound Dendrobium Officinale(CDO) combined with Irbesartan and Amlodipine in spontaneously hypertensive rats(SHR), and to investigate the potential mechanism. Methods: SHR rats of 2 months old in Experiment 1 were randomly divided into model group, CDO group, Irbesartan group, Irbesartan and Amlodipine group(IA group), CDO combined with Irbesartan and Amlodipine group(CDOIA group), and Wistar-Kyoto(WKY) rats were taken as normal group, with 10 rats in each group, orally administered for 6 weeks. SHR rats of 8 months old in Experiment 2 were randomly divided into model group, IA group, and CDOIA group, with 6 rats in each group, and 5 WKY rats were taken as normal group, orally administered for 4 weeks. Systolic blood pressure(SBP) was measured in each group of rats at 3 h and 24 h after administration. Serum angiotensin Ⅱ(AngⅡ) and insulin were measured by enzyme-linked immunosorbent assay. The relative mRNA expressions of AngⅡ type 1 receptor(AT1R), AngⅡ type 2 receptor(AT2R), insulin receptor(ISR), insulin α receptor(ISR-α), and insulin β receptor(ISR-β) were estimated by Real-time fluorescence quantitative PCR of heart. Results: At 3 h and 24 h after administration during the whole treatment process, SBP in CDOIA group was significantly lower than that in the IA group(P<0.01);the serum AngⅡ in CDOIA group was significantly higher than that in model group(P<0.05);the mRNA expression of AT1R in IA group and CDOIA group was significantly higher than that in model group(P<0.05);the mRNA expression of ISR in CDOIA group was significantly higher than that in IA group(P<0.05). Conclusion: Compound Dendrobium Officinale combined with Irbesartan and Amlodipine has significant antihypertensive and synergistic effects, and its mechanism may be related to the up-regulation of ISR mRNA expression.
