摘  要:目的:建立不同批次生/制何首乌蒽醌部位的UPLC指纹图谱,结合模式识别与含量测定对其进行质量评价和鉴别,并考察其抗氧化活性。方法:采用UPLC对35批生/制何首乌蒽醌部位进行检测,建立UPLC指纹图谱,运用相似度分析、聚类分析、偏最小二乘-判别分析对其进行评价,确认生/制何首乌分类的主要贡献峰,并对主要差异性化学成分进行含量测定;采用ABTS法测定35批生/制何首乌蒽醌部位的抗氧化活性。结果:建立生/制何首乌蒽醌部位的UPLC指纹图谱,标定6个共有峰,其中2个峰分别被指认为大黄素和大黄素甲醚;通过聚类分析和偏最小二乘-判别分析基本能区分生/制何首乌,并确定大黄素为生/制何首乌的主要差异性化学成分;生/制何首乌蒽醌部位中大黄素的含量分别为99.07~199.40、36.44~98.09 mg/g,ABTS自由基清除率分别为75.07%~85.93%、53.36%~74.93%,其抗氧化活性与大黄素含量可能存在一定的关系。结论:本研究可为生/制何首乌质量整体信息控制、鉴别和抗氧化活性提供试验依据。Objective: To identify and evaluate its quality and antioxidant activity of anthraquinone part in crude and processed Polygonum multiflorum from different producing areas by UPLC fingerprints combined with pattern recognition and content determination. Methods: UPLC was used to detect 35 batches of anthraquinone part in crude and processed Polygonum multiflorum, and the UPLC fingerprints were established. Similarity analysis, cluster analysis and partial least squares-discrimination analysis were used to evaluate and find the main contribution peaks leading to the classification of crude and processed Polygonum multiflorum, and content determination of main different chemical components was conducted. ABTS method was used to determine antioxidant activity. Results: UPLC fingerprints of anthraquinone part in crude and processed Polygonum multiflorum were established, and 6 common peaks were marked, 2 of which were referred to as emodin and physcion. Cluster analysis and partial least squares-discriminant analysis could basically separate crude and processed Polygonum multiflorum, and emodin were determined as the main different chemical components between crude and processed Polygonum multiflorum. The contents of emodin of anthraquinone part in crude and processed Polygonum multiflorum were 99.07~199.40,36.44~98.09 mg/g, respectively, and the eliminating rates of ABTS free radical were 75.07%~85.93%, 53.36%~74.93%,respectively. The eliminating rates of ABTS free radical were related to the contents of emodin. Conclusion: Fingerprints combined with pattern recognition and content determination can be used to identify and evaluate its quality and antioxidant activity of anthraquinone part in crude and processed Polygonum multiflorum.
