摘 要:目的:观察滋补脾阴方药对脾阴虚大鼠胃肠激素、水液代谢及线粒体能量代谢的影响。方法:将SPF级雄性Wistar大鼠随机分为正常组(C组)、脾阴虚组(S组)和滋补脾阴方药干预组(Z组),利用饮食失常兼过劳结合燥热耗伤阴液法建立脾阴虚大鼠模型。采用ELISA法检测胃窦组织中胆囊收缩素(CCK)和空肠组织中血管活性肠肽(VIP)的表达水平。采用免疫组织化学染色法观察空肠组织中水通道蛋白4(AQP4)的表达水平。采用Western-blot法测定胃、空肠与胰腺组织中与能量代谢相关的信号通路分子的表达水平。结果:与C组大鼠比较,S组大鼠的胃窦组织CCK水平及空肠组织VIP水平显著上调(P<0.05),滋补脾阴方药干预后能显著降低其水平(P<0.01)。免疫组化染色结果显示,脾阴虚大鼠空肠中AQP4的表达水平显著升高(P<0.01),滋补脾阴方药干预后能显著降低AQP4的表达水平(P<0.05)。Western-blot结果显示,与C组比较,S组空肠CaM、CaMKⅡ、p-CaMKⅡ蛋白表达水平显著升高(P<0.01,P<0.05);胰腺p-Akt蛋白水平、胃ClpP蛋白酶、Lon蛋白酶表达水平显著降低(P<0.01,P<0.05)。给予滋补脾阴方药干预后,上述蛋白的表达水平均显著改善(P<0.01,P<0.05)。结论:脾阴虚使大鼠胃肠激素、水液代谢及线粒体能量代谢产生显著改变。滋补脾阴方药能够显著改善脾阴虚大鼠的上述改变,可能是其发挥对脾阴虚证干预作用的机制之一。Objective: To observe the effects of Zibu Piyin Recipe(ZBPYR) on gastrointestinal hormone, water metabolism and energy metabolism in spleen-yin deficiency model rats. Methods: SPF male Wistar rats were randomly divided into normal group(group C), spleen-yin deficiency group(group S) and ZBPYR group(group Z). The spleen-yin deficiency rat model was replicated by eating disorder and overfatigue combined with dryness and heat depletion of yin liquid. The expression levels of cholecystokinin(CCK) in stomach and vasoactive intestinal peptide(VIP) in the intestine were detected by ELISA assay and the expression level of aquaporin 4(AQP4) in intestinal tract was observed by histochemical staining. The expression levels of energy metabolism-related signaling pathway molecules in stomach, intestine and pancreas were determined by Westernblot method. Results: Compared with group C, CCK and VIP levels in groups was significantly up-regulated(P<0.05). After the intervention of ZBPYR, CCK and VIP levels was significantly decreased(P<0.01). Immunohistochemical staining results showed that the expression level of AQP4 in intestinal tract of rats with spleen-yin deficiency was significantly increased(P<0.01), and the expression level of AQP4 could be significantly decreased after the intervention of ZBPYR(P<0.05). Western-blot results showed that the protein expression levels of CaM, CaMKⅡ and p-CaMKⅡ in jejunum in group S were significantly increased compared with the group C(P<0.01, P<0.05). Pancreatic p-Akt protein level, the expression levels of ClpP and Lon protein in stomach were significantly decreased(P<0.01, P<0.05). After the intervention of ZBPYR, the expression levels of these proteins were significantly improved(P<0.01, P<0.05). Conclusion: The gastrointestinal hormones, water and fluid metabolism and mitochondrial energy metabolism in spleen-yin deficiency rats are significantly changed.
