摘  要:目的:观察熊胆粉及熊去氧胆酸对原发免疫性血小板减少症(ITP)模型小鼠骨髓和循环血Ly6C^(hi)单核细胞分布及相关细胞因子含量的影响,探讨其对ITP的作用机制,为蒙药熊胆替代品研究提供依据。方法:利用豚鼠抗小鼠血小板血清(APS)复制/建立ITP模型,将小鼠随机分为正常组、模型组、泼尼松组、熊胆组和熊去氧胆酸组,治疗10 d。流式细胞仪检测骨髓和循环血Ly6C;单核细胞分布,酶联免疫吸附(ELISA)测定血清肿瘤坏死因子-α(TNF-α)和白介素-1β(IL-1β)含量。结果:与模型组比较,熊胆组和熊去氧胆酸组骨髓Ly6C^(hi)单核细胞表达率显著升高(P<0.05),而循环血Ly6C;单核细胞表达率显著减少(P<0.05),外周血TNF-α和IL-1β含量显著减少(P<0.05)。结论:熊胆粉及熊去氧胆酸可通过抑制Ly6C^(hi)单核细胞从骨髓进入循环血,改善炎症因子的分泌,从而发挥治疗ITP作用。Objective: To observe the effects of bear bile and ursodeoxycholic acid(UDCA) on circulating blood, bone marrow Ly6C^(hi)monocyte and serum related cytokines in mice with immune thrombocytopenia(ITP), and to explore its action, to provide a reference for studying bear bile and its succedaneum treated on ITP. Methods: As is the previously study, to establish ITP model by using the method of immunization, mice were randomly divided into normal group, model group, prednisone group,bear bile group and UDCA group, after treatment 10 d, the changes of the Ly6C^(hi)monocyte in circulating blood and bone marrow were detected by flow cytometry, the levels of serum tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β) were examined by Enzyme-linked immunosorbent Assay. Results: After treatment, compared with the model group, the percentage of Ly6C^(hi)monocyte subsets in bone marrow were obviously higher in bear bile group and UDCA group(P<0.05), while the percentage of Ly6C^(hi)monocyte subsets in circulating blood were lower(P<0.05), the levels of TNF-α and IL-1β decreased(P<0.05). Conclusion: The bear bile and UDCA can treat ITP by inhibiting the egress of Ly6C^(hi)monocyte emigration from bone marrow to circulating blood and improving the secretion of inflammatory factors.
