摘  要:多糖蛋白结合疫苗的研发及成功上市是预防细菌性传染病(bacterial infectious diseases)的突破,目前已批准上市的多糖蛋白结合疫苗主要有b型流感嗜血杆菌结合疫苗、肺炎球菌结合疫苗和脑膜炎球菌结合疫苗。载体蛋白的选择是多糖蛋白结合疫苗的免疫原性增强的关键因素,目前获批上市的疫苗主要使用的载体蛋白有破伤风类毒素(tetanus toxoid,TT)、白喉类毒素(diphtheria toxoid,DT)和白喉毒素突变体(CRM;)、不可分型流感嗜血杆菌(non-typeable Haemophilus influenzae,NTHi)蛋白D以及重组铜绿假单胞菌菌外毒素A(recombinant exotoxin A of Pseudomonas aeruginosa,rEPA)等。与此同时,多价疫苗的研发以及多种疫苗联合接种已成为当前细菌性疫苗发展的方向,但不同载体蛋白的结合疫苗同时接种可能引起免疫干扰。现对多价疫苗和联合疫苗接种时其相关的免疫应答影响作一概述,旨在为今后多糖蛋白结合疫苗的研发方向以及相关接种程序提供借鉴或启示。The development and successful marketing of polysaccharide-protein conjugate vaccines is a breakthrough for the prevention to bacterial infectious diseases.Currently,the licensed polysaccharide protein conjugate vaccines mainly include Haemophilus influenzae type b conjugate vaccine,Pneumococcal conjugate vaccine and Meningococcal conjugate vaccine.The selection of carrier protein is a key factor in polysaccharide-protein conjugate vaccine preparation,which the carrier proteins mainly used would be tetanus toxoid(TT),diphtheria toxoid(DT)and cross-reacting material 197(CRM;),as well as non-typeable Haemophilus influenzae(NTHi)protein D or recombinant exotoxin A of Pseudomonas aeruginosa(rEPA),et al.Meanwhile,the development of multivalent conjugate vaccines and multiple conjugate vaccines co-administration also become the current research focus,while simultaneous immunization with conjugate vaccines bearing different carrier proteins might induce immune interference.This review summarizes the effect of carrier proteins to polysaccharide-induced immune responses,which aims at providing a solution or enlightenment for the future research and related vaccination procedures.
