摘  要:为获得大黄芩鱼散半仿生提取物的制备工艺,本研究以均匀实验设计U5(53)安排实验,考察提取溶媒pH、料液比、提取时间对提取物抗鹅沙门氏菌最小抑菌浓度(MIC)的影响,以乙腈-0.1%磷酸水为流动相梯度洗脱,高效液相色谱法检测提取物中的活性成分。结果显示:大黄芩鱼散半仿生提取最优的工艺为pH 8.3,料液比1:40,提取时间0.5 h,对鹅沙门氏菌的MIC为(9.10±0.11)mg/mL,HPLC可以同步检测提取物中的活性成分有黄芩苷、槲皮素、芦荟大黄素和大黄素。The purpose of this study was to obtain the preparation technology of semi-biomimetic extract on Dahuangqinyu powder. The effects of solvent pH,solid-liquid ratio and extraction time on the minimum inhibitory concentration(MIC) of the extracts against Salmonella goose were investigated by the table U5(53) of uniform experimental design. The active components in the extracts were detected by high performance liquid chromatography(HPLC) after gradient elution using acetonitrile-0.1% phosphoric acid water as mobile phase.The results showed that the optimal semi-bionic extraction conditions were solvent pH 8.3,solid-liquid ratio 1:40,extraction time 0.5 h,MIC of this extracts against Salmonella goose was(9.10±0.11)mg/mL,and baicalin,queretin,aloe emodin and emodin in the extract could be simultaneously detected by HPLC.
