异源CD151蛋白在H9c2细胞中的表达对PRRSV感染的影响

摘　　要：猪繁殖与呼吸综合征(porcine reproductive and respiratory syndrome,PRRS)是由PRRS病毒(PRRS virus,PRRSV)引起的以猪繁殖障碍和呼吸系统症状为特征的一种急性、高度传染的病毒性疾病。研究表明PRRSV可在表达CD151的一株鼠源细胞系中复制增殖,提示异源CD151可能与PRRSV复制增殖相关。为了进一步验证异源CD151本文运用是否参与病毒的复制增殖,本研究验证了表达CD151的大鼠来源的心肌细胞系H9c2对PRRSV的易感性。RT-PCR和间接免疫荧光分析(indirect immunofluorescence analysis,IFA)表明H9c2细胞系中表达CD151蛋白和波形蛋白。病毒吸附试验和攻毒试验提示该病毒可以与H9c2细胞结合,但接种病毒后细胞培养上清和细胞裂解液中均不能检测到病毒核衣壳N蛋白表达,表明病毒不能在该细胞中复制。这一结果提示H9c2细胞中表达的异源CD151不能支持PRRSV的复制增殖。Porcine reproductive and respiratory syndrome(PRRS)is an acute,highly contagious viral infection disease caused by PRRS virus(PRRSV),which is characterized by reproductive disorders and respiratory symptoms in pigs.Previous studies have shown that virus-sensitive cells expressing CD151 are associated with replication of PRRSV.In this study,RT-PCR and indirect immunofluorescence analysis(IFA)studies have showed that PRRSV can express CD151 protein in murine H9c2 cells.Virus adherent and challenge experiments showed that the virus could bind to H9c2 cells,but no nucleocapsid N protein expression could be detected in cell culture supernatant and cell lysate,indicating that the virus could not replicate in this cell.These results indicated that murine H9c2 cells could not be used as PRRSV proliferating cell line.
