摘  要:目的研究凉血活血方与益气活血方对不同时期放射性肺损伤大鼠不同部位血流情况的影响及两方剂所含成分分析。方法80只SD大鼠,随机分成4组:照射组、照射+凉血活血组、照射+益气活血组和空白对照组,每组20只。除空白组外,其余各组大鼠以6 MV-X线经背部照射右肺,单次3 Gy,每周照射2次,连续照射5周,最高累积剂量30 Gy,建立放射性肺损伤模型;于照射开始时,凉血活血组和益气活血组同步灌胃给药,每日一次;模型组和空白组给予同等体积生理盐水;分别于照射开始后第12周、第26周对大鼠各部位(耳背、背侧胸部、腹侧胸部及腹部)的血流情况进行采集。通过液质联用技术对凉血活血和益气活血方药中所含成分进行解析鉴定。结果与空白对照组比较,在放射性肺损伤急性期,模型组大鼠背侧胸部及腹侧胸部血流量均显著升高(P<0.01)。在背侧胸部,与模型组相比,凉血活血组和益气活血组急性期大鼠的血流量均显著降低(P<0.01),且凉血活血组效果优于益气活血组(P<0.01);在腹侧胸部,与模型组相比,凉血活血组急性期大鼠腹侧胸部血流量显著降低(P<0.01),耳背部和腹侧腹部各组间无明显统计学差异。液质联用技术鉴定表明,在mzCloud数据库中,凉血活血方共匹配到564个化合物,254个化合物综合评分大于70;益气活血方共匹配到188个化合物,139个化合物综合评分大于70。在两方中,评分大于92分且共有的成分是汉黄芩素、18-β-甘草次酸、洋川芎内酯H、麦芽四糖、齐墩果酸等成分。结论凉血活血方和益气活血方均可改善急性放射性肺损伤大鼠背侧胸部的血流变化,且凉血活血方更具有一定优势;通过液质联用技术可进一步分析鉴定两方药所含成分的区别,为临床运用及进一步研究提供参考。Objective To investigate the effects of Liangxue Huoxue prescription and Yiqi Huoxue prescription on the blood flow in different parts of radiation-induced lung injury rats and analyze the components in the two prescriptions. Methods Eighty SD rats were randomly divided into four groups: irradiation group, irradiation+Liangxue Huoxue group, irradiation+Yiqi Huoxue group and blank control group, with 20 rats in each group. Except for blank control group, the rats were irradiated with 6 MV-X ray to establish a radiation-induced lung injury model through the back to the right lung for a single 3 Gy irradiation, twice a week for 5 consecutive weeks, with a maximum cumulative dose of 30 Gy. At the beginning of irradiation, the rats in Liangxue Huoxue group and Yiqi Huoxue group were synchronized intragastric administration once a day. The rats in irradiation group and blank control group were given the same volume of saline once a day. The blood flow volume of the part of the rat(back of the ear, dorsal chest, ventral chest and abdomen) was collected at week 12 and week 26 after the irradiation. In addition, the ingredients contained in two prescriptions were analyzed and identified by liquid chromatography-mass spectrometry technology. Results Compared with blank control group, the blood flow volume in the dorsal chest and the ventral chest were significantly increased in the acute phase of radiation-induced lung injury in irradiation group(P<0.01). In the dorsal chest, compared with model group, the blood flow volume in the acute phase was significantly reduced in Liangxue Huoxue group and Yiqi Huoxue group(P<0.01), and the effect in Liangxue Huoxue group was better than in Yiqi Huoxue group(P<0.01). In the ventral chest, compared with model group, the blood flow volume of rats in acute phase was significantly reduced in Liangxue Huoxue group(P<0.01), and there was no statistically significant difference in the back of the ear and the ventral abdomen between four groups.
