摘　　要：目的 观察miR-30a-3p在复发性流产患者绒毛组织中的表达,及其对滋养细胞(HTR-8)凋亡、迁移功能的影响。方法 采用qRT-PCR检测复发性流产患者(RSA组)及正常早孕妊娠(正常组)要求终止的患者各20例绒毛组织中miR-30a-3p的表达水平。在HTR-8细胞中转染miR-30a-3p前体分子,共分3组:miR-30a-3p模拟物组(mimic组)、miR-30a-3p抑制物组(inhibitor组)和阴性对照组(NC组)。流式细胞术检测细胞凋亡,Transwell试验检测细胞侵袭能力。结果 qRT-PCR结果显示,与正常组相比,RSA组绒毛组织中miR-30a-3p表达量显著降低(P<0.05)。与NC组相比,mimic组HTR-8细胞中miR-30a-3p表达升高,inhibitor组miR-30a-3p表达水平降低(P<0.05)。流式细胞结果显示,与NC组相比,inhibitor组细胞凋亡率显著增加(P<0.05)。Transwell结果显示,与NC组相比,inhibitor组迁移能力显著降低(P<0.05)。结论 复发性流产绒毛组织中低表达的miR-30a-3p可能导致滋养细胞凋亡增加,迁移力降低,从而导致复发性流产的发生。Objective To investigate the expression of miR-30 a-3 p in chorionic villus tissues of patients with recurrent abortions, and its effect on the apoptosis and migration of trophoblast(HTR-8) cells. Methods RT-qPCR was used to detect the expression level of miR-30 a-3 p in the villus tissues of 20 patients with recurrent miscarriage(RSA group) and 20 patients who required termination of normal early pregnancy(normal group). The miR-30 a-3 p precursor molecules were transfected into HTR-8 cells, and divided into three groups: miR-30 a-3 p mimic group(mimic group), miR-30 a-3 p inhibitor group(inhibitor group), and negative control group(NC group). Flow cytometry was used to detect the apoptosis in each group, and Transwell method was used to detect the invasion ability in each group. Results The qRT-PCR results showed that the expression of miR-30 a-3 p in RSA group was significantly lower than that in normal group(P<0.05). Compared with NC group, the expression of miR-30 a-3 p was increased in HTR-8 cells in mimic group, and decreased in inhibitor group. The flow cytometry results showed that compared with NC group, the apoptosis rate in inhibitor group was significantly increased(P<0.05). Transwell results showed that compared with NC group, the migration ability was significantly reduced in inhibitor group(P<0.05). Conclusion Low expression of miR-30 a-3 p in villous tissue of recurrent miscarriage may lead to the increase of trophoblast cell apoptosis and the decrease of migration, which leads to the occurrence of recurrent miscarriage.
