摘 要:为进一步利用血液资源,以羊血清蛋白为原料来探究胰蛋白酶水解羊血清蛋白的工艺.以水解度为指标,试验采用单因素研究pH、温度、加酶量和水解时间四个因素对水解度的影响.在此基础上,采用Box-Benhnken响应面试验设计优化胰蛋白酶水解藏系绵羊血清蛋白的工艺条件.结果表明:在底物浓度为5%,加酶量为9000 U/g时,最佳水解条件为:反应时间5.6 h、温度55℃、pH10.0,在此条件下水解度可达到19.32±0.21%.In order to make further use of blood resources,the process of pantetic protease hydrolysis of amniotic serum protein is explored with amniotic serum protein as raw material.Taking hydrolysis degree as the index,the single factor design was used in the present experiment so as to study the effects of pH,temperature,enzyme amount and hydrolysis time on hydrolysis of serum protein.On this basis,the process conditions of panteinase hydrolysis serum protein in sheep were optimized by Box-Benhnken response surface test.The results showed that when the substrate concentration was 5%and the enzymatic amount was 9000 U/g,the optimum hydrolysis conditions were:reaction time was 5.6 h,temperature was 55℃,pH was 10.0,and hydrolysis could reach 19.32±0.21%under this condition.
