补中益气汤含药血清干预TGF-β介导的A549/DDP细胞E-cadherin、N-cadherin表达的影响

摘　　要：目的观察补中益气汤含药血清对TGF-β诱导前后,A549/DDP细胞E-cadherin、N-cadherin表达的影响,探讨补中益气汤干扰上皮-间充质转化(epithelial-mesenchymal transition,EMT)分子机制。方法细胞分为A549/DDP组;TGF-β1组(5ng/L,48h);中药组(10%补中益气汤含药血清),采用免疫细胞化学法、Western blot法、实时荧光定量PCR法检测补中益气汤含药血清对A549/DDP细胞E-cadherin、N-cadherin蛋白及mRNA水平的影响。结果与A549/DDP组比较,TGF-β1组E-cadherin蛋白及mRNA表达水平下调(P<0.05),与TGF-β_(1)组比较,中药组E-cadherin蛋白及mRNA表达水平上调(P<0.05),差异具有统计学意义。而N-cadherin蛋白及mRNA水平的影响呈现相反的变化趋势。结论E-cadherin、N-cadherin可作为EMT的重要分子标记物,且补中益气汤含药血清可下调E-cadherin蛋白及mRNA表达,上调N-cadherin蛋白及mRNA表达,逆转A549/DDP细胞EMT,进而干预肺腺癌细胞的侵袭、转移。Objective To observe the expression of E-cadherin and N-cadherin in cisplatin-resistant cell line A549/DDP mediated by transforming growth factor-β1(TGF-β1),and to explore the association of Buzhong Yiqi Decoction with epithelial-mesenchymal transition(EMT).Methods The cells were divided into A549/DDP group;TGF-β1 group(5 ng/L,48 h);Traditional Chinese medicine group(10% Serum Containing Buzhong Yiqi Decoction).The protein expression of E-cadherin and N-cadherin was determined by the methods of immumofluorescence and Western blot.The mRNA expression of E-cadherin and N-cadherin was determined by the methods of real-time PCR.Results Compared with A549/DDP group,the protein and mRNA expression of E-cadherin in TGF-β_(1) group was significantly decreased(P<0.05),and N-cadherin was significantly increased(P<0.05).Compared with TGF-β_(1) group,the protein and mRNA expression of E-cadherin in Traditional Chinese medicine group was significantly increased(P<0.05),and N-cadherin was significantly decreased(P<0.05).Conclusion E-cadherin and N-cadherin were very important molecular marker of EMT.Serum Containing Buzhong Yiqi Decoction can up-regulate the protein and mRNA expression on E-cadherin of A549/DDP cell mediated by TGF-β1 and down-regulate the protein and mRNA expression on N-cadherin,reserve the invasion and metastasis and reverse the EMT.
