摘 要:目的探究樟帮特色炮制饮片鳖血制柴胡中五种皂苷成分(SSa、SSd、SSc、SSb;、SSb;)含量的变化规律,为扩大鳖血制柴胡的临床应用提供理论支持。同时有利于推动具有区域特色中药饮片炮制的科学内涵研究。方法采集不同产地,不同来源,不同品种的13批生柴胡(北柴胡、南柴胡、锥叶柴胡),按照柴胡∶鳖血=100∶15的比例,在约110℃炒干。利用HPLC法测定五种皂苷含量,采用Agilent ZORBAX C18色谱柱,柱温30℃,流动相为乙腈-水(0.05%磷酸)梯度洗脱,流速为1.0 ml·min^(-1),检测波长分别为210nm(SSa、SSd、SSc)、254nm(SSb_(2)、SSb_(1))。结果经鳖血炮制后,所有柴胡SSa与SSd含量均有升高,增加率为10%~60%,其中道地产区陕西、山西、甘肃SSa、SSd皂苷变化量最大。陕西柴胡的SSb_(1)及山西柴胡的SSb_(2)含量略有减少,河北柴胡的SSc、SSb_(1)含量减少,其他成分含量均有升高。内蒙古柴胡五种皂苷成分含量均有升高。结论柴胡经过鳖血炮制后,五种主要皂苷成分的含量有不同程度升高和降低,但柴胡主要皂苷成分SSa、SSd有明显提高。此研究说明鳖血柴胡炮制工艺能够有效提高部分柴胡皂苷含量,为樟帮特色炮制品鳖血柴胡的应用及推广奠定了理论基础,也从现代科学的角度解释了鳖血制柴胡可挫缓柴胡"劫肝阴"之性。Objective To explore the variation rule of the contents of five saponins(SSa,SSd,SSc,SSb;,SSb;)in the special processing of turtle blood to make Bupleurum,and to provide theoretical support for expanding the clinical application of turtle blood to make Bupleurum.At the same time,it is beneficial to promote the scientific connotation research of the processing of slices with regional characteristics.Methods 13 batches of raw Bupleurum(Bupleurum chinense DC.Bupleurum scorzonerifolium Wild.Bupleurum bicaule Helm)were collected from different habitats,different sources and different varieties and dried at about 110℃ according to the ratio of Bupleurum:Trionyx blood=100:15.HPLC method was used to determine the contents of five saponins.Agilent Zorbax C18 column was used.The column temperature was 30℃,and the mobile phase consisted of acetonitrile-water(0.05% phosphoric acid)gradient elution at the flow rate of 1.0 mL·min^(-1).The detection wavelength was 210 nm(SSa,SSd,SSc),254 nm(SSb_(2),SSb_(1)),respectively.Results After processing with soft-shelled teronyx blood,the contents of SSa and SSd in all Bupleurum were increased by 10%~60%,and the changes of SSd saponins in SSa from Shaanxi,Shanxi and Gansu were the largest.The content of SSb_(1) in Bupleurum Shaanxi and SSb_(2) in Bupleurum Shanxi decreased slightly while the contents of SSc and SSb_(1) in Bupleurum Hebei were decreased,while the contents of other components were increased.The contents of five saponins in Bupleurum were all increased in Inner Mongolia.Conclusion Bupleurum after turtle blood up five main content of saponin constituents have different degree rise and fall,but the bupleurum saponins main composition SSa,SSd,has increased significantly,the study shows that turtle blood Bupleurum concocted technology can effectively improve the part of bupleurum saponins content,It laid a theoretical foundation for the application and promotion of the special processed product of turtle blood to make Bupleurum,It is also explained from the point of view
